Objectives :1.The expression and biological effects of FBXO39 in gastric cancer cell lines.Methods:1.The expression of FBXO39 was measured by western blotting and immunofluorescence in gastric cancer cells MGC-803,SGC-7901 and normal gastric epithelial cell GES-1.2.The proliferation and migration of SGC-7901 cells were measured by MTT,clone formation and cell scratch assay after FBXO39 overexpression.3.The cell cycle of SGC-7901 cells were measured by Flow cytology after FBXO39 overexpression.Results :1.FBXO39 expression level in gastric cancer cells was higher than normal cell.Western blotting and immunofluorescence used to analyze gastric cancer cells MGC-803,SGC-7901 and normal gastric epithelial cell GES-1.the experimental results finding FBXO39 expression level in gastric cancer cells was higher than normal cell,and it may play a vital role in gastric cancer.2.FBXO39 overexpression could promote SGC-7901 cell proliferation.MTT results showed that: The relative appreciation rate of FBXO39 overexpression group and control group is about 147%-182%,higher than the other two groups.the Result tell to us: SGC7901 cell proliferation was enhanced after transfection p EGFP-FBXO39.Simultaneously,we examine the SGC7901 clone-ability by clone formation.The results suggested that FBXO39 overexpression's colony forming rate was23.7±0.9,which is significantly higher than the control group 9.6 ±0.6?empty vector group 8.7 ±0.5,and PEGFP-FBXO39? F group 7.7±0.5(P<0.05).Taken together,these findings indicate that FBXO39 overexpression could promote SGC-7901 cell proliferation.3.FBXO39 overexpression could promote SGC-7901 cell migration.In order to get better to understanding of the effect of FBXO39 on SGC-7901 cell migration.We used cell scratch test,and the result was suggested that: FBXO39 overexpression group 24-hour migration rate about 69.7%,and 48-hour migration rate about 90.2%.But other three group24-hour migration rate about 45-47%,8-hour migration rate about58-61%.Results analyzed by one-way ANAOVA showed that FBXO39 group is significantly higher than others group,also tell us FBXO39 overexpression could promote SGC-7901 cell migration.4.FBXO39 overexpression could not significant effect on SGC-7901 cell cycle.Flow cytometry displayed,About the FBXO39 group,the G0/G1?S?G2/M phase of SGC-7901 cells were 47.42±2.29?35.91±1.87?35.91±1.87,compared to another groups of cells cycle is not difference(P>0.05).Conclusions:1.FBXO39 expression level in gastric cancer cells was higher than normal cell.2.FBXO39 overexpression could promote SGC-7901 cell proliferation and migration.3.FBXO39 overexpression could not significant effect on SGC-7901 cell cycle.
