The Experssion And Significance Of Smad4 And TWIST In Gastric Adenocarcinoma

Objective: To detect the relative quantitative expression of Smad4 and TWIST in gastric adenocarcinoma,paracancerous tissues and normal gastric tissues,to analyze the relationship between Smad4 and TWIST expression and its relationship with clinicopathological parameters,and to explore new biological indicators and therapeutic targets for diagnosis and treatment of gastric cancer.Methods:(1)30 patients with gastric adenocarcinoma diagnosed as gastric adenocarcinoma without radiochemotherapy were collected.30 healthy volunteers were collected to obtain tissue samples;(2)Smad4 and TWIST were detected by immunohistochemistry(SP)method.The expression of gastric cancer,paracancerous and normal gastric mucosal tissues,and the relationship between the clinicopathological parameters of gastric cancer and their expression were analyzed.Results:(1)The positive rates of Smad4 in gastric cancer tissues,paracancerous tissues,and normal tissues were: 26.67%(8/30),56.67%(17/30),63.33%(19/30),respectively.There was a statistically significant difference between paracancerous and normal tissues(P=0.035,P=0.009).There was no statistical difference between normal gastric tissues and adjacent tissues(P=0.792).(2)Expression of Smad4 in gastric cancer tissues Lymphatic metastasis,differentiation,and TNM staging were related(P<0.05);it was not related to gender,age,tumor size,tumor location,and invasion depth(P>0.05).(3)The expression rates of TWIST in gastric cancer tissues,paracancerous tissues,and normal gastric tissues were 53.33%(16/30),10.00%(3/30),3.33%(1/30),respectively.Gastric cancer tissues and paracancerous tissues Compared with normal gastric tissues,there were statistically significant differences(P=0.001,P<0.001).There was no statistical difference between adjacent tissues and normal gastric tissues(P=0.162).(4)Expression of TWIST in gastric cancer It was related to lymph node metastasis and TNM staging(P<0.05).It had nothing to do with gender,age,tumor size,tumor location,invasion depth and differentiation(P>0.05).(5)Smad4 was negatively correlated with TWIST expression in gastric cancer tissues(r=-0.451,P = 0.023).Conclusions:(1)The expression of Smad4 in gastric cancer tissues maybe low,while TWIST is highly expressed.The expression of Smad4 is negatively correlated with that of TWIST.(2)Both Smad4 and TWIST are associated with lymphatic metastasis and TNM staging of gastric cancer;Smad4 is related to the differentiation grade of gastric cancer.
