Objective To investigate the effect of SCD1 on invasion and proliferation of human breast cancer MDA-MB-231 cell.Methods1.SCD1-siRNA1,2,3 are targeting three different sites of SCD1 gene.SCD1-siRNA1,SCD1-siRNA2,SCD1-siRNA3 were transfected into human breast cancer MDA-MB-231 cell by liposomes transient transfection method as siRNA1 group,siRNA2 group,siRNA3 group.SCD1-siRNA empty transfection group was taken as an negative control group(NC group).The protein expression of SCD1 in each group was detected by Western blot(WB).2.The proliferation ability of the cell was tested by MTT test,the metastasis ability of the cell tested by cell scratch test and the invasion ability of the cell tested by Transwell invasion test.3.The protein expression of epithelial tissue-associated mesenchyme(EMT)protein in human breast cancer MDA-MB-231 cells were detected by WB.ResultsWestern Blot results showed that the expression level of SCD1 protein in the SCD1-siRNA1,SCD1-siRNA2,SCD1-siRNA3 group are significantly lower than NC group(P<0.05).The MTT test showed that the proliferation ability of siRNA groups is inhibited(P < 0.05).The wound healing assay showed that the migration ability of siRNA groups cells is significantly inhibited(P < 0.05).The number of siRNA group cells that pass through the Transwell Chambers are significantly less than NC group(P < 0.05),invasive ability is inhibited.The results of WB test showed that inhibiting the expression of SCD1 increased the expression of E-cadherin,reduced the expression of N-cadherin and ?-catenin(P < 0.05),SCD1 promoted the EMT transformation of human breast cancer cell MDA-MB-231.Conclusions1.SCD1 increase the invasion,proliferation and metastasis ability of human breast cancer MDA-MB-231 cell in vitro.2.SCD1 effect the EMT transformation of human breast cancer cell MDA-MB-231.
