Heshouwu,the raw root of Plygonum multiflorum Thunb,is one of the most popular traditional Chinese medicines and is an ingredient in many medicines and prescriptions.Plygonum multiflorum is considered as tonic medicines for hair-blacking,liver-nourishing,kidney-nourishing,hematopoiesis and so on.However,an increasing number of recently published studies have demonstrated the adverse effects of Polygonum multiflorum.Some researchers have found that Polygonum multiflorum shows not only hepatotoxicity but also a possible drug interaction.Specifically,the long-term use of Polygonum multiflorum may lead to liver and kidney toxicity.The main component s(2,3,5,4’-Tetrahydroxystilbene-2-O-glc and emodin)of Polygonum multiflorum are poorly absorbed in vivo,and their blood drug concentration and bioavailability arelow in vivo.First,the objective of the previous study was to develop a practical strategy for the identification of metabolites following the in vivo metabolism and in vitro microbial biotransformation of Polygonum multiflorum extract using LTQ-Orbitrap XL.As a result,101 compounds(15 parent drugs and 86 metabolites)were characterised in rat urine,plasma,bile,faeces and intestinal after the oral administration of Polygonum multiflorum extract.Nine metabolites and 6 parent drugs of 2,3,5,4’-Tetrahydroxystilbene-2-O-glc and anthraquinone were isolated from the urine and faeces of rats after the oral administration of Poly gonum multiflorum extract,and their structures were elucidated on the basis of spectroscopic data.The metabolites were characterized as citreoro sein(M-1),acid-citreorosein(M-2),citreorosein-1-O-sulfate(M-3),emodin-1-O-sulfate(M-4),emodin-1-O-glucuronide-8-O-sulfate(M-5),emodin-1,6-O-glucuronide(M-6),emodin-1,8-O-glucuronide(M-7),3,5,4’-hydrogetetrahydroxystilbene-3-O-glucuronide-4’-sulfate(M-8),3,5,4’-hydrogetetrahydroxy stilbene-4’-sulfate(M-9).The parent drugs were characterized as 2,3,5,4’-Tetrahydroxystilbene-2-O-glc(M-10),Emodin(M-11);Physcion(M-12),Chr ysophanic acid(M-13),Emodin-O-hex(M-14),Aloe-emodin(M-15).8 metabolites were new compounds(M-2,M-3,M-4,M-5,M-6,M-7,M-8,M-9),7 comp ounds(M-1,M-10,M-11,M-12,M-13,M-14,M-15)were isolated for the first time from urine and faeces after the oral administration of Polygonum multiflorum extract.The metabolites of main components of Polygonum multiflorum were characterized,which is important to understand its metabolic fate and disposition in humans.
