| Objective:To explore the effects of benzoylmesaconine(BMA)on Proliferation and Apoptosis of Human Multiple Myeloma Cell RPMI8226(RPMI8226).Methods:Logarithmic growth RPMI8226 cells were randomly divided into blank control group and BMA(13,65,130,1300,13000 ng · mL-1,)groups,CCK8 was used to determine the effect of BMA on the activity of RPMI8226 cell line,estimated the IC50 based on the results:According to the IC50,we randomly divided Logarithmic growth RPMI8226 cells into lank control group,negative control group and BMA(520,780,1040 ng·mL-1)groups,Changes of proliferation inhibition rate of BMA on RPMI8226 cell line with concentration and time was determined by CCK8;According to the rate of proliferation,we randomly divided Logarithmic growth RPMI8226 cells into lank control group and BMA(520,1040 ng·mL-1)groups,the effect of BMA on RPMI8226 apoptosis was detected by AnnexinV/PI double staining flow cytometry.Resulis:Within the range of 13ng ·mL-1,tol3000ng ·mL-1 of BMA for 24 hours,the activity of RPMI8226 cells decrease with the increase of concentration(P<0.05),the IC50 is about 1040ng mL-1.the proliferation inhibition rate of RPMI8226 increases with the increase of concentration and touch time of BMA,all P values<0.05,the difference is statistically significant.Apoptosis rate of RPMI8226 cells increases with the increase of concentration and touch time of BMA,all P values<0.05,the difference is statistically significant.Conclusions:BMA can inhibit the proliferation of RPMI8226 and induce the apoptosis of the cell line. |
