Purpose:The effects of Qige Tongluo prescription on diabetic peripheral neuropathy in rats were observed and compared in body weight,blood glucose,histopathologic changes of sciatic nerve,nerve conduction velocity,HMGB1 protein and mRNA expression of sciatic nerve.In order to provide theoretical basis for clinical treatment,the clinical curative ef fect and the mechanism and approach of its treatment are discussed.Method:The rat model of diabetic peripheral neuropathy was induced by streptozotocin(STZ).After the model was successfully established,the rats were randomly divided into normal control group,model group,metformin group(140mg·kg-1·d-1),Qige Tongluo high-dose group(7.36g·kg-1·d-1),Qige Tongluo low-dose group(1.84g·kg-1·d-1),each treatment group was given corresponding medication by gavage.After 12 weeks of intervention,blood glucose,body weight,histopathologic changes of sciatic nerve,nerve conduction velocity,HMGB1 protein and mRNA expression in sciatic nerve were detected.Result:After treatment:1)The treatment of body weight and blood glucose of rats significantly improved compared with the model group(P<0.05).2)Compared with the model group,the MNCV and SNCV were increased in all the treatment groups(P<0.05),and the Qige Tongluo high-dose group was significantly improved.3)The morphology of sciatic nerve in the model group was obviously changed,and the morphology of sciatic nerve in the treatment groups was improved after the intervention of Qige Tongluo prescription.The improvement was especially obvious in the high dose group of Qige Tongluo prescription(P<0.05).4)The expression of HMGB1 protein and mRNA in the high dose group,low dose group and metformin group were decreased.There was significant difference between the model group and the model group(P<0.01).Conclusion:The Qige Tongluo prescription has a good effect on the prevention and treatment of diabetic peripheral neuropathy.Its possible mechanism is to inhibit inflammation and apoptosis by decreasing blood sugar,down regulating the expression of HMGB1 protein and mRNA,so as to prevent and control Diabetic nephropathy.
