Background: Systemic lupus erythematosus(SLE)is an autoimmune disease which usually occurs in women of childbearing age.Its clinical manifestations are extensively heterogeneous,which skin,joints,kidneys and central nervous system display inflammation accompanied by antinuclear antibodies produced at the same time.The pathogenesis of SLE may be related to the immune maladjustment of T cells and B cells,where B cells are hyperactive,producing autoantibodies.In addition to environmental factors,the development of SLE contains an important genetic factor.SLE is regarded as a kind of complex genetic diseases and the most recent GWAS reported a number of susceptibility genes,including interleukin 10(IL 10)gene with association of SLE among Asian and African populations.T-cell activation GTPase activating protein(TAGAP)is involved in T lymphocyte activation.Recently,several literatures have reported that TAGAP gene is associated with the genetic susceptibility of some other autoimmune diseases such as rheumatoid arthritis(AS)in European,African and Chinese populations,but there is no research to explore the gene for the genetic risk of SLE.Therefore,this study was undertaken to investigate the association between mononucleotide polymorphisms of TL-10 and TAGAP genes and SLE in Yunnan Chinese population.Methods: the 500 normal control samples and 500 cases of systemic lupus erythematosus were all from the second affiliated hospital of Kunming medical college.Genotyping of four SNPs(rs1800890,rs1800872,rs1738084 and rs21389)was performed using nested PCR-RFLP,and SPASS17.0 was used for statistical analysis.Results: Logistic regression analysis showed that genotype CC of rs1800872 of IL-10 gene was significantly different(P=0.023),and was protective factor of SLE(OR=0.465).In the analysis of gender stratification,there was significant difference in the female genotype CC(P=0.038;OR=0.444).SNP rs1738074 genotype of the TAGAP gene had significant difference(P=0.02)and was protective factor of SLE(OR=0.696).The allele G has significant difference(P=0.007,OR=0.79),which was protective factor of SLE.In the analysis of gender stratification,there was a significant difference between the female genotype GG and the allele G(P<0.05).There was no association of IL-10 gene rs1800890 and TAGAP gene rs212389 with the development of SLE.Conclusion: IL-10 gene rs1800872 and TAGAP gene rs1738074 may be related to the development of SLE in Yunnan Chinese population.
