Primary liver cancer is one of the most common malignancies and its main pathological type is hepatocellular carcinoma(HCC).At present,the treatment of HCC is still a comprehensive method based on surgical resection,The related statistics shows that the 5-year overall survival rate is still very low,and 60%-80% of patients recur in 2 years after operation.Postoperative recurrence especially early recurrence is the most serious problem that influences the prognosis of HCC after hepatectomy.But there is still lacking in effective predictors for early recurrence of HCC.Consulting the documents and materials,we know that the expression of SERPINB3 increases in chronic liver diseases,dysplastic nodules and HCC,high expression of SERPINB3 in liver tumours is significantly associated with early tumour recurrence.Objective:1.To compare the expression of SERPINB3 mRNA levels in tissues with that in adjacent non-tumor tissues of HCC patients2.To investigate the correlation of SERPINB3 mRNA levels with clinical outcome of HCC patients3.To explore the independent risk factors for early recurrence of HCC after curative resection.Materials and Methods:The clinical data and tissue samples of 60 eligible patients who underwent surgical resection in No.1 Hospital of Jilin University from 2012-4 to 2016-1 were collected.Real-time quantitative PCR was used to detect the expression levels of SERPINB3 in HCC tissues and adjacent non-tumor tissues.To explore whether the expression of SERPINB3 mRNA levels in HCC tissues was noticeablely increased compared with adjacent non-tumor tissues and to investigate the correlation of SERPINB3 mRNA levels with clinical outcome of HCC patients.The patients were divided into early recurrence group(<2 years)and no early recurrence group by recurrence time.The clinicopathological factors of the two groups were analyzed in order to find the related factors influencing the early recurrence of HCC after curative resection Results:1.The expression of SERPINB3 mRNA levels in HCC tissues was noticeablely increased compared with adjacent non-tumor tissues(P=0.001)2.Correlation of SERPINB3 expression with clinicopathological features was significant only for the presence of TNM stage(P=0.01)and microvascular invasion(P=0.017)3.Results of univariate analysis indicted that TNM stage?Tumor size(>5cm)?microvascular invasion?HBV-DNA(>1000IU/ml)?over-expression of SERPINB3(>1.9714)were associated with early recurrence,But multivariate Logistic regression analysis indicted that tumor size(>5cm)? HBV-DNA(>1000IU/ml)? over-expression of SERPINB3(>1.9714)were independent risk factors for early recurrence.Conclusions1.The expression of SERPINB3 in HCC tissues was noticeablely increased compared with adjacent non-tumor tissues.2.Tumor size(>5cm)?HBV-DNA(>1000IU/ml)and overexpression of SERPINB3(>1.9714)were independent risk factors for early recurrence.3.The overexpression of SERPINB3 was associated with TNM stage and microvascular invasion.
