Objective: To study the expression level of SOX4 gene in patients with acute myeloid leukemia(AML)and its correlation with clinical characteristics and prognosis,and to explore its role and clinical significance in acute myeloid leukemia.Methods:Real-time quantitative PCR was used to detect the expression of SOX4 gene in bone marrow leukocytes in 96 patients with primary AML and 26 controls.The correlation between the expression of SOX4 gene and clinical characteristics and prognosis was analyzed.Results: SOX4 expression in 96 AML patients was significantly higher than that in controls(P = 0.001).There was no significant difference in SOX4 gene expression between M1-M5 patients(P = 0.258).There was no significant difference in the expression level of SOX4 gene between different genders and nationalities and clinical characteristics such as remission after one course of chemotherapy(P>0.05).The level of SOX4 in AML patients was not significantly correlated with the laboratory indexes such as leucocyte count,hemoglobin,platelet count,blasts count and reticulocyte count(P>0.05).The high expression of SOX4 influenced the overall survival of patients Rates of OS(P = 0.003)and event-free survival EFS(P = 0.023)were independent prognostic factors.COX multifactorial analysis found that the proportion of blast cells,WT1,SOX4 gene expression is an independent factor affecting OS(P = 0.014,P = 0.001,P = 0.003),EFS(P = 0.032,P = 0.001,P = 0.024),While the results also showed that age is the factor that affects EFS(P = 0.025).Conclusion: The SOX4 gene is highly expressed in the bone marrow of patients with acute myeloid leukemia and is one of the unfavorable prognostic factors affecting patient survival(OS,EFS).
