Objective:Screening the proteins that predict the Chemotherapy efficacy and prognosis in Esophageal Squamous Cell Carcinoma,and in esophageal squamous cell carcinoma cell lines to identify its function.Methods:1.Inclusion of 52 cases of advanced and locally advanced ESCC patients,the standard of radiotherapy and chemotherapy,to determine the efficacy and long-term follow-up,while collecting patients before and after treatment of serum.2.Using isotope labeled relative and absolute quantification(iTRAQ)technology to screen the differential therapeutic effect and prognosis related proteins in radiotherapy and chemotherapy.3.After RNAi lentiv irus was used to infect experimental cell lines,the expression of ILK gene was ab olished.The effects of ILK gene ablation on cell proliferation and apoptosis were determined by Celigo,MTT and FACS.Results:1.Short-term efficacy evaluation: 59.62%(31/52)of patients with esophageal cancer(CR+PR+SD)and radiotherapy and chemotherapy(PD)accounted for 40.38%(21/52).Follow-up results of long-term follow-up results: survival time of 1-2 years group accounted for 46.15%(24/52 cases);survival period 2-3 years group 32.69%(17/52 cases);survival period ? 3years group accounted for 21.15%(11/52 cases).2.Esophageal cancer patients before radiotherapy and chemotherapy than the normal healthy control group was up-regulated(r = 4.386,p <0.05);radiotherapy and chemotherapy sensitive group than radiotherapy and chemotherapy antagonist group was down-regulated(r = 0.587,p <0.05);?3 years of survival was reduced in the 1-2 year group(r=0.633,p<0.05).3.Screening for IL-1 highly expressed esophageal squamous carcinoma cell line TE-1;after infection with lentivirus,the expression level of ILK mRNA at the mRNA level in experimental group TE-1 cells was inhibited(p<0.05),knockdown efficiency reached 88%.After inhibiting the expression of ILK gene,the number of cells that have undergone apoptosis was found to increase in the cell line,the proliferation of esophageal cancer cells slowed,and the number of esophageal cancer cell clones decreased.Conclusion:ILK has the potential value of predicting the efficacy and prognosis of esophageal cancer radiotherapy and chemotherapy;inhibition of ILK gene can inhibit the proliferation of esophageal squamous cell carcinoma cells and promote their apoptosis.
