Objective: Study on Osteogenic Differentiation of Rabbit Periosteal Cells and Bone Marrow Mesenchymal Stem Cells under Different Conditions under Intermittent Mechanical Centrifugal Stress Stimulation.Methods: The rabbit periosteal and femoral bone marrow were cultured in vitro under aseptic conditions.The periosteal cells were continuously cultured and the bone marrow mesenchymal stem cells were transferred to the third generation and inoculated on six-well plates.The principle of randomization was divided into plus group and blank group,centrifugal force loading on the force group with a bench-top low-speed centrifuge.After the centrifugal force was adjusted to a corresponding size,the force group was loaded with stress stimuli of 170 g,210 g and 250 g.The control group received no stimulation and the rest of the conditions were the same for the ultra-clean bench at 10 minutes per day.Samples were taken periodically to determine the ALP activity of the cells,the proliferation ability of each group of cells was determined by MTT assay,and staining was detected for mineralization.Real-time fluorescence quantitative PCR was used to determine the relative expression levels of osteocalcin-related genes osteocalcin OCN and RUNX2 mRNA in both cells.Results: 1.ALP activity increased and proliferative activity increased after periosteal cells and BMSCs were loaded by intermittent mechanical centrifugal force at 170 g,210g and 250 g.2.The proliferative activity,ALP activity,number of mineralized nodules,and the relative expression of osteocalcin OCN and RUNX2 mRNA were significantly increased in the 250 g periosteum group and the 170 and 250 g groups of BMSCs(P<0.05).Conclusion: 1.Intermittent mechanical centrifugal force stimulation can promote the proliferation and mineralization of rabbit periosteum cells and BMSCs and promote the expression of OCN and RUNX2.2.Under the same conditions,170 g,210g,and 250 g of the two cells in the two groups of cells,250 g periosteum group,170 g and 250 g group of BMSCs cell proliferation activity,ALP activity,number of calcified nodules,osteocalcin OCN and RUNX2 mRNA relative The amount of expression has increased significantly.3.The osteogenic effect of rabbit periosteum cells and BMSCs is related to the size of intermittent mechanical centrifugal force.
