Purpose:To investigate the radiosensitization effect of Bosutinib on hepatocellular carcinoma cell Hep G2 and its possible mechanism,and to provide experimental basis for the clinical improvement of radiotherapy of liver cancer.Method:The trials were designed as simple drug group,single radiotherapy group(2Gy),single radiotherapy group(4Gy)drug + radiotherapy group(2Gy)and drug + radiotherapy group(4Gy);using CCK-8 assay,two time points were selected(24h 48h)Detecting the inhibitory effect of different drug concentrations and different radiation doses on human hepatoma cells Hep G2;further cell cloning experiments to observe cell survival.Finally,the cell cycle of each group was examined by flow cytometry.Statistical data SPSS22.0 was used to process the data.The multivariate correlation method and variance analysis method were used to analyze the inhibitory effects of various factors on human hepatoma cells Hep G2 and the roles among various factors.Result:1.The growth inhibition rate of hepatoma cells Hep G2 in single drug group was 1000 n M/ml and 600 n M/ml after 24 h and 48 h respectively.There was a time-dose effect on the cell survival fraction.Analysis of variance showed that there was a statistically significant difference in drug concentration and action time when the drug concentration was greater than 200 n M/ml(P<0.05).The radiotherapy dose and bosutinib concentration all inhibited the growth of human hepatoma cells Hep G2,and the drug concentration and radiotherapy dose had a synergistic effect.By analyzing the inhibitory rate of hepatoma cells and the correlation of bosutinib concentration,time,radiotherapy dose,the results showed that the growth inhibition rate was affected by the dose of radiotherapy,bosuritib concentration,and the effect of time,and there was a positive correlation.The data results were The analysis was statistically significant(P<0.05).The results of the experiment suggest that bosutinib has a significant effect on the growth inhibitory rate of hepatocellular carcinoma cell line Hep G2,and shows a sensitizing effect in the in vitro culture experiment of hepatocellular carcinoma cells,and the sensitizing effect is more pronounced with the dose of radiotherapy.2.In the cell cloning experiment,when the concentration of bosutinib was set to 100 n M/ml,bosutinib alone combined with radiotherapy had a strong inhibitory effect on the growth and proliferation of human hepatoma cells Hep G2,with the radiation dose With the increase,bosurtinib combined with radiotherapy has a more pronounced inhibitory effect on the proliferation of Hep G2 cells.3.After 24 hours and 48 hours of treatment with different drug concentrations,the cell cycle distribution was detected.It was found that the proportions of G1,G2,and S phase cells changed with the increase of drug concentration and time of action.With the increasing concentration of bosutinib,the percentage of the cell cycle in the S phase of the cell cycle is gradually reduced within 24 hours of the effect,and the proportion of the cell cycle in the S phase decreases after 48 hours.more obvious.After bosutinib combined with radiotherapy,the percentage of each cell cycle was examined,and the results were statistically significant after analysis(P<0.05).The distribution of the cell cycle of liver cancer changes more significantly.Conclusion:Bosutinib can significantly inhibit the growth of hepatoma cells,and bosuritib combined with radiotherapy has a more pronounced inhibitory effect on the proliferation of Hep G2 cells.Bosutinib has a strong radiosensitization effect,and its mechanism may be achieved by changing the phase distribution of the cell cycle.
