Objective:In this study,the changes of plasma miR-302 c and HSP90 alpha levels in patients with non-small cell lung cancer(NSCLC)before and after chemotherapy and before and after drug resistance were analyzed,respectively analysis the correlation between the expression of the plasma miR-302 c,HSP90 alpha and chemotherapy drug resistance,To investigate the clinical value of the expression of plase miR-302 c and HSP90 alpha in chemoresistance of lung cancer.Methods:1.A total of 60 patients with confirmed NSCLC who had not received any treatment were collected at the first affiliated Hospital of Nanhua University from January 2016 to September 2017.We respectively collect their limosis venous blood 5ml before chemotherapy and when they are treated with chemotherapy(Unify to take after the 2 cycles of chemotherapy and the patients who have been evaluated for CR/PR).Finally,a total of 30 cases patient's limosis venous blood of chemotherapeutic resistance(SD/PD)were collected in the follow-up of 60 patients.2.Plasma miRNAs were extracted from 90 plasma samples,and c DNAs were synthesized by reverse transcription,and then real-time fluorescence quantitative polymerase chain reaction(PCR)was used to detect miR-302 c expression in patients with lung cancer.Corresponding plasma samples were tested for HSP90 alpha expression by enzyme-linked immunosorbent assay(ELISA).3.SPSS 22 statistical software was used to analyze the experimental data.Single factor analysis of variance was used to compare the three groups:the pre-chemotherapy group,the non-resistance group after chemotherapy and the resistance group.Chi-square test was used to analyze the relationship between the expression of plasma miRNA-302 c and clinical pathological features,such as sex,age,smoking history,differentiation,clinical stage,lymph node metastasis and so on.The independent sample t test was used to analyze the correlation between the expression of HSP90 alpha and the clinical pathological features.According to the three groups expression results of plasma miRNA-302 c,HSP90 ?,the average was calculated relative expression,analysis the changes of plasma miR-302 c and HSP90? expression in patients with lung cancer before and after chemotherapy and before and after drug resistance,evaluation of plasma miR302-c,HSP90? in lung cancer chemotherapy curative effect evaluation and forecast chemotherapy drug resistance in the clinical application value.Results:1.The expression level of plasma miR-302 c in NSCLC patients with chemotherapeutic resistance was significantly higher than that of non-drug-resistant patients after chemotherapy.(P < 0.05).2.The expression level of plasma miR-302 c in NSCLC patients before chemotherapy was higher than that of non-drug-resistant patients after chemotherapy,but the difference without Statistical significance.(P> 0.05).3.The expression level of plasma HSP90? in NSCLC patients with chemotherapeutic resistance was significantly higher than that of non-drug-resistant patients after chemotherapy.(P < 0.05).4.The expression level of plasma HSP90? in NSCLC patients before chemotherapy was significantly higher than that of non-drug-resistant patients after chemotherapy(P < 0.05).5.The overexpression of miRNA-302 c in plasma was correlated with lymph node metastasis(P < 0.05)and distant metastasis(P < 0.05)in NSCLC.The high expression of HSP90? was associated with the distant metastasis of non-small cell lung cancer(P < 0.05)There was no significant correlation with gender,age,degree of differentiation,clinical stage,smoking history,etc.in patients with lung cancer(p>0.05).6.The expression of miR302 c was found to be positive correlated with HSP90? in NSCLC(rs=0.423,P=0.02).Conclusion:1.The high expression of plasma miRNA-302 c and HSP90? is associated with chemotherapy resistance in non-small cell lung cancer.2.Plasma miRNA-302 c and HSP90? may be one of the markers of sensitivity of chemotherapy drugs.3.The expression of plasma miRNA-302 c was up-regulated after chemotherapy resistance and was positively correlated with the expression of HSP90?.
