Objective: To investigate the effect of Paeonol on the proliferation,migration,invasion and the MAPKs signaling in human lung cancer A549 cells.Methods: CCK-8 method was used to investigate the inhibitory efficacy of different concentrations of Paeonol on human lung cancer A549 cells,and IC50 was calculated.A549 cells were randomly divided into four groups,namely the blank control group,Paeonol 6.25 ?g/m L group,Paeonol 12.5 ?g/m L group,Paeonol 25 ?g/m L group.Scratch assay and Transwell assay were performed to investigate the action of Paeonol on the migration and invasion characteristic of A549 cells.The expression level of MMP-2 and MMP-9 proteins on the culture supernatant were detected by Elisa assay.Alteration of MAPKs signaling in A549 cells under the influence of Paeonol was detected by the western blot assay and the phosphorylation levels of p-ERK1/2,p-JNK and p-P38 proteins were monitored.Results Paeonol could inhibit the proliferation of human lung cancer A549 cells in a concentration dependent manner,the IC50 was 54.68±3.59 ?g/m L.Cell scratch tests confirmed that different concentrations of phenol(6.25 ?g/m L,12.5 ?g/m L,25 ?g/m L)could inhibit the migration ability of A549 cell in a concentration dependent manner,with the increase of Paeonol concentrations,the 24 h scratch healing rates gradually decreased when compared with the blank control group(P< 0.05).Results of Transwell invasion experiment show that Paeonol could significantly suppress the invasion ability of A549 cells.With the concentration increased,the number of cells penetrated the membranes reduced gradually,and the differences were statistically significant(P< 0.05)when compared with blank control group(0 ?gg/m L).Elisa assay showed that the expression level of MMP-2 and MMP-9 in the cultural supernatants of A549 cells could be reduced by 6.25 ?g/m L,12.5 ?g/m L and 25 ?g/m L of Paeonol,and the differences were statistically significant(P< 0.05)when compared with blank control group(0 ?g/m L).The results of western blot revealed that Paeonol could suppress the phosphorylation level of ERK1/2 signaling in MAPKs compared with the blank control group(P< 0.05),while it did not show inhibitory effect on p38 and JNK signaling.Conclusion In a certain concentration range,Paeonol could inhibit the proliferation,migration and invasion characteristics of human non-small cell lung cancer A549 cells,and suppress the secretion of MMP-2 and MMP-9 proteins,these actions may attribute to the inhibitory effect of Paeonol on the ERK1/2 signaling in MAPKs of A549 cells.
