Synthesis,Biological Activity And Application Of Asymmetry Curcumin Analogues

17 compounds were synthesized in this study,12 of which are asymmetric analogues and 10 of which have never been reported in literature.All of the compounds' structures were synthesized and evaluated by IR?H1NMR?LC-MS.We expect to see that asymmetric single carbonyl curcumin analogues also have good stability,antioxidant activity and anti-tumor activity.In this paper,we used acetone as the carbon line to synthesized 17single carbonyl curcumin analogues.Altering th e carbon chain of curcumin from seven carbons to five carbons could improve its stability.And the 4-OH group was more stability than 2-OH.All of the compounds performed better in an anti-proliferate assay than curcumin,especially compound 25,which exhibited the preferential cytotoxic activity against MCF-7 cells(25,IC50=9.11 ?M,curcumin,IC50=70.2 ?M).Most compounds exhibited stronger antioxidant activity than VC,and the antioxidant activity of compound 14 which have the same substituent groups with Curcumin exhibited a stronger activity than curcumin against human hepatoma cell line(SMMC-7721),the human breast cancer cell line(MCF-7),and the human prostate cancer cell lines(PC-3).In vitro free radical scavenging experience,we evaluated four free radicals:1,1-diphenyl-2-picryl-hydrazyl(DPPH),2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS),ROO·(TRAP)and O2-.(NBT)assays.All synthetic compounds,except compound 12 and 13,performed good antioxidant activity.We selected compound 14 and 25 added into the fish oil to determined the oxidative stability of fish oil stored for 70 days at 25 ? and 4 ?.The investigation focused on the increase in acid(AV),peroxide(PV),conjugated diene(CD)values,thiobarbituric acid-reactive substances(TBARS)as well as polyunsaturated fatty acid(PUFA).The temperature is a key condition for fish oil storage,and the antioxidant activity of antioxidants are also associated with temperature.Shorten the carbon line can improve its stability,antioxidant activity and antitumor activity.4-OH is the key part of the antioxidant activity.
