Juglans mandshurica is one of the ancient and important economic tree species,which is one of the precious "three hardened" tree species in the northeast of China,belong to Tertiary species.Juglans is Originated in western Asia,now distributed throughout the world and more than 40 countries.From the domestic market,in view of the nutrition of Juglans mandshurica,health care value and better economic,social and ecological benefits,in the fruit industry structural adjustment more and more consumers and producers of all ages.In this paper,the yield and total reducing power of Juglans mandshurica were used as raw materials.The single factor test and the response surface method were used to optimize and fit the regression model.Two methods were proposed for the extraction of thermostatic water bath and microwave To compare,to obtain the best extraction methods and process conditions.The results showed that the optimum extraction conditions were as follows:the ratio of material to liquid 1:24,ethanol concentration 57%,extraction temperature 55?,extraction time 3h,the total flavonoid yield was 8.74mg/G,the empirical test,the relative standard deviation of 1.54%.The optimum extraction conditions were as follows:the ethanol concentration was 55%,the ratio of material to liquid was 1:34,the microwave power was 640W and the microwave time was 13min.The yield of total flavonoids was 9.59mg/g under the optimum conditions.The relative standard deviation was 1.98%.The total flavonoid yield and total reducing power were higher than that of the total temperature,and the total flavonoid content and the total reducing power were not changed after 7 h after extraction.The regression equation is in good agreement with the actual situation,and it is suitable for extracting flavonoids from walnut.In order to study the safety and antitumor activity of flavonoids from Juglans mandshurica,acute toxicity test was carried out.On the proliferation of HepG-2,HepG-2B,N87 and MB-231 cells(MTT test)and the effect on tumor cell morphology.The results showed that the flavonoids of Juglans mandshurica were extracted by ultrasonic wave and purified by HPD-600 macroporous resin.The inhibitory effect on the proliferation of HepG-2 cells was significantly increased with the increase of flavonoid concentration The highest inhibitory rate was as high as 62.33%at the concentration of 1000?g/ml.The morphological changes of the tumor cells can be inhibited by the inverted microscope,and the number of tumor cells can be reduced and the morphological changes can obviously inhibit the growth of these four kinds of tumor cells,and the degeneration of the tumor cells is changed.There is a dose-effect relationship.
