Purpose:The traditional Chinese medicine licorice was taken as the research object,and the active ingredient glycyrrhizic acid was extracted.The extraction conditions were optimized by response surface methodology.And using glycyrrhetinic acid as raw material,combined with computer-aided drug design and molecular modeling to carry out structural transformation research,synthesis of glycyrrhizic acid derivatives,reduce the side effects of pseudo-aldosteronism.Method:1.Through single-factor experiments,the response surface methodology was designed to study the extraction conditions of anti-virus active ingredients of glycyrrhizic acid,and the optimal extraction conditions were selected based on the yield of glycyrrhizic acid.2.Using glycyrrhetinic acid as raw material,the carboxyl group at position 30 in the structure was esterified by synthetic chemistry.3.In structural identification,thin layer identification,UV absorption spectrum identification,infrared absorption spectrum identification,and H1 nuclear magnetic resonance spectrum identification were mainly performed.4.Combining computer-aided drug design methods,the molecular structure of the synthesized product was analyzed and the experimental results were verified on the structure.Result:1.The optimal extraction conditions of glycyrrhizic acid,an effective anti-virus component of traditional Chinese medicine,were: ammonia concentration 0.8%,ethanol concentration 35%,p H value of acid sink 1.55;2.The five compounds synthesized from glycyrrhetinic acid as raw materials are glycyrrhetinic acid methyl ester,n-propyl glycyrrhetinate,isopropyl glycyrrhetinate and butyl glycyrrhetate.The structure of the resultant product was determined.3.The results of molecular structural analysis of synthetic products by computer-aided drug design were basically in agreement with experimentally measured data.Conclusion: The results of the molecular structure analysis of the synthetic products by computer-aided drug design are basically in agreement with experimentally measured data.Compared with glycyrrhizic acid,its derivatives can theoretically extend the half-life in vivo,and structural transformation is expected to enhance the drug properties and reduce the side effects of human body.This study provides theoretical and experimental evidence for the design of innovative drugs based on glycyrrhizic acid,and strives to make new breakthroughs in drug modification and efficacy improvement of glycyrrhizic acid and its derivatives,in order to provide drug synthesis and design of antiviral components for Chinese medicine.Research provides examples.
