Purpose:To investigate and study the pufall resources in Jilin Province,to clarify the material basis of pufall's pharmacodynamics,to provide basis for the improvement of the quality standards of the pufall,and to the development and utilization of the pufall resources.Method:In This study,we investigated the resources of pufall in Jilin Province,and identified the specimens of pufall we collected by the methods such as basal origin,traits,microscopic and physical and chemical methods.The strain was purified by single spore culture method and tissue separation method.The strain was fermented by liquid deep fermentation technology to obtain the fermented products of puffball.Using organic solvent extraction,LC,TLC,HPLC and other chromatographic methods to separate the chemical components of the fruit body and mycelium of Mycenastrum corium?Guers.?Desv.UV,1HNMR,13CNMR,ESI-MS and other methods were used to identify the separated chemical components.Thedeterminationofergosta-5,7,22-triene-3?-oland ergosta-4,6,8?14?,22-tetraen-3-one in the major ingredients of pharmacokinetics in Jilin Province was performed by HPLC dual wavelength method.Pharmacological experiments were conducted to study the therapeutic effects of ethanol extract of Geastrum triplex Fisch on LPS-induced acute lung injury in mice and the therapeutic effect of ergosterol on renal injury induced by streptozo-tocin in diabetic mice.Result:In this study,more than 500 specimens of pufall were collected and 25 species of 7 genera,4 families,were identified.10 strains of P.barbadensis were isolated and purified.Six compounds were isolated and identified from the fruit body and mycelium of Mycenastrum corium?Guers.?Desv.The content of ergosta-5,7,22-triene-3?-ol and ergosta-4,6,8?14?,22-tetraen-3-one in 16 kinds of puffins was determined.The results of pharmacological experiments showed that the ethanol extract of Geastrum triplex Fisch could significantly reduce the serum levels of TNF-?,IL-6,IL-1?,SOD and MDA in mice with LPS-induced acute lung injury.Ergosterol can significantly reduce the induction of streptozotocin.The level of triglyceride?TG?,total cholesterol?TC?,uric acid?SUA?and creatinine?Scr?in diabetic kidney injury mice effectively inhibited the phosphorylation of PI3K,Akt,NF-?B p65 and I?B?,and improved renal tissue changes.Conclusion:The resources of pufall in Jilin Province are abundant.Most of the pufall species in China are distributed in Jilin Province.The steroid components is one of the main effective components of pufall.Among them,ergosta-5,7,22-triene-3?-ol and ergosta-4,6,8?14?,22-tetraen-3-one in steroids can be used as indicators for the qualitative identification and quantitative identification of pufalls.The fluorescent properties of spores and spores can be used as the important features of microscopic identification of puffballs.Pufall fermented material and fruiting body are similar in chemical composition and have similar pharmacological effects,and can be exploited and used as alternatives to pufall medicine.The results of pharmacological experiments showed that the ethanol extract of Geastrum triplex Fisch has a significant therapeutic effect on LPS-induced acute lung injury in mice,and ergosterol has a significant therapeutic effect on kidney injury induced by streptozotocin in diabetic mice.
