OBJECTIVE: Rab1 A expression in colorectal malignancies was investigated by laboratory methods.The clinical significance of Rab1 A protein expression in the diagnosis and prognosis of colorectal cancer patients was determined by statistical software,which further laid the foundation for the diagnosis and treatment of colorectal cancer.METHODS: A total of 100 cases of human colorectal tissues and 29 cases normal colorectal tissues that were more than 5 cm away from the edge of tumor were obtained from October 2009 to April 2014 in the department of general surgery,the second affiliated hospital of Soochow university.We initiated our investigation by immunohistochemistry and western blot analysis to confirm Rab1 A expression in CRC tissues.Meanwhile,the correlation of Rab1 A expression and clinicpathological parameters as well as outcome in CRC patients was examined,and to explore the relationship between Rab1 A expression and the prognosis of patients with colorectal cancer.RESULTS: In the issue,Rab1 A is overexpressed in CRC tissues compared with normal colorectal tissues(P<0.001).Meanwhile,high Rab1 A expression was significantly associated with TNM stage(P=0.010),lymph node metastasis(P=0.037)and peritoneal metastasis(P=0.032).In addition,multivariate analyses suggested that Rab1 A expression(P=0.047)and TNM stage(P=0.013)served as an independent predictor for CRC patients.Moreover,Kaplan-Meier survival analysis showed that patients with high Rab1 A expression had a significantly worse survival time than those with low Rab1 A expression(P=0.001).CONCLUSIONS: Compared with normal colorectal tissue,the expression of Rab1 A protein in colorectal malignant tumor tissue was high.These results suggest that Rab1 A is an important diagnostic marker for CRC,and Rab1 A can be used as a valuable biomarker for prognosis in CRC patients.
