Objectives: To investigate the relationship between serum inflammatory cytokines and prognosis of patients with small cell lung cancer.Methods: 1.we retrospectively reviewed 80 newly diagnosed patients with small-cell lung cancer at the First Affiliated Hospital of Jilin University,From January 7,2014 to December 20,2016,including 48 males(60%)and 32 females(40%);the ratio of male to female was 2.0 to1,the median age were 58 years(ranged from 31 years to 79 years).2.At the time of the initial diagnosis,3 to 5 ml of venous blood was collected from the each patient.After centrifugation,50?l of serum was separated in an EP tube using a pipette and stored in a refrigerator at-80°C.The levels of 17 Inflammatory Factors(IL-1?,IL-2?4? 5?6?7?8?10? 13,IL-12p70,IL-17 A,G-CSF,GM-CSF,IFN-?,MCP-1,MIP-1?,and TNF-?)were measured by Luminex x MAP multiplex assays(Luminex),which have been widely used in other biomarker studies.Then we should collecte the basic clinical data of the patients and calculate the progression-free-survival(PFS),overall survival time(Overall survival,OS).3.The statistical software package SPSS version 17.0(SPSS Inc.,Chicago,IL,USA)was used for statistical analysis.All categorical data were expressed as n(%)and analyzed using Fisher's exact probability test.OS and progression-free survival(PFS)curves were plotted using the Kaplan-Meier method and analyzed using the log-rank sum test.Multivariate analysis using Cox proportional hazards model.The hazard ratio(HR)was expressed as the 95% confidence interval(CI).P-value less than 0.05 was considered statistically significant.Results: 1?IL-5 high water group(n=28)and low level(n=52)were found to have similar characteristics in terms of gender,age,smoking history,Eastern Cooperative Oncology Group(ECOG)performance statusand clinical staging(all P-values >0.05).The median OS of IL-5 high-levels and low-levels lgroups were 11.61 months(95% confidence interval [CI],9.81-11.41 months)and 17.60 months(95%CI,14.67-19.43 months)respectively.The PFS values were 9.14 months(95% CI,7.65-10.62months)and 11.74 months(95%CI,9.24-14.25months),respectively.Both univariate and multivariate analysis suggested that the high levels of IL-5 were associated with Shorter OS correlations(P=0.017,P=0.029),while IL-5 levels were not associated with PFS.2.IL-13 high water group(n=41)and low level(n=39)had no statistical differences in gender,age,smoking history,Eastern Cooperative Oncology Group(ECOG)performance status,and clinical stage(all P-values >0.05).The median OS of IL-13high-levels and low-levels groupswere 13.63 months(95% confidence interval [CI],11.60-15.68 months)and 16.74 months(95% CI,13.88-19.60 months),respectively.The PFS values were 9.18 months(95% CI,7.74-16.62 months)and 12.57 months(95% CI,9.72-15.72months),respectively.Univariate and multivariate analysis suggested that the high levels of IL-13 were associated with Shorter OS correlations(P=0.044,P=0.044),while IL-13 levels were not associated with PFS.3.There was no statistical difference in gender,age,smoking history,Eastern Cooperative Oncology Group(ECOG)performance status,and clinical stage(all P-values >0.05),between MCP-1 high water group(n=40)and low level(n=40).The median OS of the high-and low-level MCP-1 groups was 13.35 months(95% confidence interval [CI],10.80-15.89months)and 16.95 months(95% CI,14.60-19.30months),respectively.The PFS values were 9.45 months(95% CI,7.19-11.70 months)and 12.22 months(95% CI,9.63-14.80 months),and univariate and multivariate analyses all suggested that the high levels of IL-13 were associated with The shorter OS was related(P=0.040,P=0.049),while the MCP-1 level had no correlation with PFS.4.Serum levels of IL-1?,IL-2?4?6?7?8?10,IL-12p70,IL-17 A,G-CSF,GM-CSF,IFN-?,MIP-1?,TNF-? were no significant difference in PFS and OS between lung cancers.5.The level of IL-8 was related to the stage of small cell lung cancer.The levels of IL-8 in the serum of patients with limited-stage and extensive stage small cell lung cancer were 8.91(5.39-15.09)pg/ml and 14.58(9.69-18.04)pg/ml,respectively.P=0.021 The difference was statistically significant.Conclusion: 1.High levels of IL-5,IL-13 and MCP-1 in serum are independent prognostic factors of poor OS after treatment for small cell lung cancer.2.Serum IL-8 levels are related to the severity of small cell lung cancer.
