Objective:Detected the expression of Tim-3 in patients with chronic lymphoc ytic leukemia(CLL),and to explored its clinical significance in chronic lymphocyti c leukemia.Methods:Choose between January 2016 and January 2017 in our hos pital treated,48 cases of patients treated in our hospital were enrolled in the exper imental group(group of CLL patients),and 20 healthy subjects were enrolled in th e control group(healthy adult group),The expression of Tim-3 in peripheral blood CD3~+,CD4~+and CD8~+T lymphocytes in the experimental group and the control g roup were measured by flow cytometry(FCM).The correlation of Tim-3 expressio n with beta 2-microglobulin,serum lactate dehydrogenase(LDH),clinical stage,CD38 expression,ZAP-70 expression,and the relevance of FISH detect cytogenetic abn ormalities.Results:(1)Among the 48 patients with CLL,30 male patients and 18female patients,The ratio of male to female is about 1.67:1,Age range is 40-85 y ears old,median age is 59.5 years,Compared with the control group,there was n o statistically significant difference in age and gender(P>0.05).In the various indi cators of peripheral blood,the difference between the group of patients with chroni c and healthy adults was statistically significant(P?0.05).(2)The expression of ti m-3 in peripheral blood CD3~+,CD4~+and CD8~+T lymphocytes in the patients with CLL patients was higher than that in the healthy control group.The difference bet ween the two groups was statistically significant(P?0.05).(3)In Binet staging,th e expression level of Tim-3 was gradually increased in BinetA,BinetB and BinetC.In the Rai stage,the expression level of Tim-3 increased with the increase of the risk(low,medium and high risk group),and the expression level of Tim-3 was a lso increase(P?0.05).(4)CD3~+,CD4~+and CD8~+T cell surface expression of Tim-3in CD38 positive group is higher than in the CD38 negative,positive group were higher than in the ZAP-70 negative group,the prognosis of chromosomes is highe r than in the genome,good prognosis in beta 2 microglobulin acuity 3.5 mg/L is higher than the beta 2 microglobulin<3.5 mg/L,The difference was statistically sig nificant(P?0.05).(5)The expression of tim-3 was not correlated with serum lactat e dehydrogenase(LDH),and the difference was not statistically significant(P>0.05).Conclusion:This study reveals that Tim-3 were highly expressed in CLL patients.Its expression levels were correlated with the period of clinical disease,CD38,ZA P70 expression,abnormal chromosome karyotype and?2-MG,but not with LDH.Ti m-3 may be indicators of prognosis in patients with CLL.
