Objective: To investigate the expression of PIK3 CA and PTEN in PI3K/Akt/mTOR signaling pathway in diffuse large B cell lymphoma and their clinicopathological correlation.Method :A total of 205 fresh cases of diffuse large B cell lymphoma were collected from January 1st 2008 to December 31 st 2012 from Xinjiang First Affiliated Hospital of Xinjiang Medical University.Ten cases of lymph node reactive hyperplasia were collected as contrast.Samples were fixed by dehydration and other pre-treatment paraffin-embedded sections,and then two pathologists complete histological diagnosisbased on morphology and immunohistochemical phenotype.Using Gene Technology's PIK3 CA and PTEN probes and LEICA situ hybridization instrument to complete the experiment,the application of SPSS 17.0 software for statistical analysis.Results: The overall rate of PIK3 CA expansion in diffuse large B-cell lymphoma was 12.7%(26/205).The positive rates of PIK3 CA in stage I-II and III-IV of clinical stage of DLBCL were(10 / 123,8.1%;16 / 82,19.5%)higher than the former(P =0.016)Statistical significance.While the overall deletion rate of PTEN was 15.1%(31/205)in diffuse large B cell lymphoma,which was not correlated with other clinicopathological features.There was a negative correlation between PIK3 CA amplification and PTEN deletion(P = 0.044).There was no significant correlation between PIK3 CA amplification and PTEN deletion and survival.Conclusions: The amplification rate of PIK3 CA in stage ?-? of DLBCL is higher than PIK3 CA.The amplification of PIK3 CA is negatively correlated with the deletion of PTEN,which suggests that PIK3 CA may promote the occurrence of DLBCL.
