Lycorine is belong to Lycoris plants.The study found many kinds of pharmacological effects of Lycorine,such as anti-tumor effect,anti-virus effect and anti-inflammatory effect and so on.Therefore,the development and utilization of lycorine can provide more help for the cure and medical treatment of related diseases.Its future is very promising and it will be a new research hotspot.This experiment is about the study on Lycorine's retardant mechanism on the humanSGC-7901 cell cycle by the in vitro test.To provide the theoretical foundation for the antitumor cycle arrest mechanism of lycorine and the theoretical basis for the intensive study on utilization of antitumor drugs and their thera peutic methods.In this experiment,the MTT method was used to test that the growth inhibition of lycorine on SGC-7901 cells.The results showed that the half inhibition rate(IC50)was 36.48?mol·L-1 on SGC-7901 cells,72h after dosing.The research found that lycorine had significant growth inhibition effect on SGC-7901 cells.With the increasing of Lycorisidine dose,the growth inhibition of rate of gastric cancer SGC-7901 cells also increased graduallyFlow cytometry was used to detect the cell cycle of human gastric cancer cell line SGC-7901 and the effect of theophylline on the cell cycle of human gastric cancer cell line SGC-7901 was inhibited.In the blank control group,the positive drug hydroxycamptothecin 9.6?mol·L-1,and after the human gastric cancer cell line SGC-7901 had been worked by three different concentrations of Lycorine(18.24?mol·L-1?36.48?mol·L-1?72.96?mol·L-1)for 48 h respectively.The result of compared with the blank control group is that G1 and the percentage of cells in S phase decreased,and the proportion of cells in G2 phase was 4.976%,49.452%,16.888%,30.244%,64.735%.With the increase of the concentration of lycorine,the intensity of block increased gradually,The relationship was appeared,there are significant differences.The results showed that Lycorine could effectively block gastric cancer cell line SGC-7901 in G2/M phase and inhibit the cell cycle of human gastric cancer cell line SGC-7901.The crystal violet staining method was used to study the effects of Lycorine on the nuclei of human gastric cancer SGC-7901 cells.the nuclei of Lymphocytes had been inhibited by Lycoriside at concentrations of 18.24?mol·L-1?36.48?mol·L-1?72.96?mol·L-1 For 48h.The results showed that there was no obvious morphological changes in the cells of the blank control group,and maintained the normal morphology of adherent growth.In the treatment group,the morphological changes of the gastric cancer SGC-7901 cells changed significantly with the increase of the concentration of the lycorine,by tight connection becomes loosely arranged,flat polygonal cells become oval,cell gap was significantly increased,part of the cells have begun to detachment of chromatin gradually concentrated,the nucleus broken.Lycorisidine gastric cancer SGC-7901 cells can cause chromosome condensation,nuclear fragmentation,and the higher concentration,the more obvious the phenomenon of cohesion;Western Blot analysis was used to detect the protein expression of Lysine(9.62?mol·L-1)After the human gastric cancer cell line SGC-7901 had been worked by three different concentrations of Lycorine(18.24?mol·L-1?36.48?mol·L-1?72.96?mol·L-1)for 48h,the CDK1/?-actin of human gastric cancer SGC-7901 cells were 0.9183,0.634,0.7881,0.7033,0.5753.The results showed that with the increase of dose,the expression of CDK1/CDK1 expression levels,medium and high dose groups have significant differences.The cell cycle related proteins CyclinB1/?-actin were 0.9862,0.7149,0.8281,0.7848,0.7099.The results showed that with the increase of dose,the expression of CyclinB1 was down-regulated,with the significant difference between middle-dose and high-dose groups.The cell cycle related proteins CDc25C/?-actin were 0.9459,0.0978,0.5761,0.3765,0.1304 respectively.The results showed that the CDc25C expression level was down-regulated with the increase of the dosage,and there was a significant difference between the low dose group,middle dose group and high dose group.Cell cycle related proteins FoxM1/?-actin were 0.8478,0.6449,0.8001,0.6562,0.5921.The results showed that FoxM1 expression was down-regulated with increasing doses,with significant differences between the medium and high doses.Tumor cells in the G2/M phase cycle play a role.Cell cycle related proteins BubR1/?-actin were 0.3638,0.1498,0.2994,0.1728,0.1094.The results showed that the expression of BubR1 was down-regulated with the increase of the dose of BubR1.There was a significant difference between the low dose group,the medium dose group and the high dose group.In summary,Lycorine can arrest human gastric cancer cell line SGC-7901 arrest in G2/M phase and cause DNA damage,broken nuclear,leading to cell apoptosis.Its mechanism is:on the one hand,by inhibiting the formation of CDK1/CyclinB1 complex,the activity of CDK1/CyclinB1 complex can be decreased by down-regulating the expression of CDc25C and reducing the activity of CDK1/CyclinB1 complex;on the other hand spindle checkpoint deletion will lead to aneuploidy The occurrence of cycle arrest caused mitotic catastrophe,BubR1 expression decreased,induction of gastric cancer SGC-7901 cells mitosis.
