The Expression And Its Clinical Significance Of LACTB In Colorectal Cancer

OBJECTIVE To research the expression of mitochondrial protein LACTB(Serine beta lactamase-like protein LACTB)in CRC(colorectal cancer)organization and colorectal normal tissues.To proved whethe the level of LACTB expressed is differement or not in colorectal cancer organization and colorectal normal tissues.To find the relation between different level of LACTB and prognosis of patients and clinical pathological characteristics.Methods(1)To selected colorectal cancer pathological paraffin blocks and source of the same cases of normal colorectal tissues adjacent to carcinoma specimens of paraffin embedding which archive of primary surgical treatment for the first time,without radiation and chemotherapy of from Qingdao municipal hospital east department January 2010 to September 2010(2)The expression of LACTB in colorectal cancer tissue and normal tissue adjacent to carcinom were detectaed by immune histochemical methods IHC(Immunohistochemistry).The degree of LACTB expressed was judged according to the immunohistochemical staining degree,and divides into high expression and low expression group according to the expression of LACTB(3)The five-year survival rate after the operation was obtained by follow-up investigation by telephone and outpatient service,thedifferences of pathological factorsand prognosis between the two groups were analyzed with the software SPSS 19(Statistical Program for Social Sciences 19).Result(1)The degree of LACTB expressed in colorectal cancer tissues and adjacent normal tissues was different,and the difference was statistically significant(p<0.01).(2)The expression level of LACTB in colorectal cancer tissues was related to the clinical pathological stages of cancer tissues(p<0.01).The higher of the clinical pathological stage,the lower of the LACTB expression.(3)The expression level of LACTB was significantly reduced in patients with lymph node metastasis compared with patients with no lymphatic metastasis,(p<0.01).(4)The 5-year survival rate of patients after the operation with low LACTB expression in cancer tissues was significantly lower than those with high LACTB expression(P=0.010).Conclusion The level of LACTB in colorectal cancer tissues was significantly lower than that in normal tissue adjacent to carcinoma,the level of LACTB had declineed accompany with the progress of tumor stage,and high degree of tumor in patients had earlier clinical generally.This prompt LACTB maybe inhibits the invasion and lymph node metastasis of colorectal cancer as a tumor suppressor.The level of LACTB is related to clinical prognosis of patients.The main risk factor for poor prognosis of cancer patients was deemed to low level of LACTB and high tumor stage.The level of LACTB is proportional to the 5 years of survival rate of postoperative patients.It can be used as markers for judging the prognosis of patients with colorectal cancerin in some extent,it also can provide new targets for the treatment of colorectal cancer as a potential tumor suppressor.
