ObjectiveTo explore the expression and diagnostic value of 6 kinds of miRNAs(mir-15b-5p,mir-17-5p,mir-19a-3p,mir-20a-3p and mir-146b-3p)in circulating blood between early non-small cell lung cancer,benign lung nodules and healthy control.Methods1.Collected by pathological diagnosis of 118 cases of non-small cell lung cancer early,42 cases with benign pulmonary nodules and 48 cases healthy crowd peripheral blood circulation.2.The total RNA was extracted from serum by QIAzol method after high speed and low temperature centrifugation.And use stem-loop primers method for reverse transcribed into cDNA,finally by RT-qPCR(real-time fluorescent quantitative PCR)to detect the case 6 kinds of miRNAs(mir-15b-5p,mir-17-5p,mir-19a-3p,mir-20a-3p and mir-146b-3p)in peripheral blood the relative expression quantity.3.SPSS19.0 was used for statistical processing: through independent sample t test,the relative expression of 6 miRNAs in each group was analyzed,and the related risk factors were identified by Logistic regression analysis and Logistic regression formula was obtained,and the diagnosis value of early non-small cell lung cancer was evaluated by the ROC Curve(Receiver Operating Characteristic Curve).Result1.Compared to patients with benign pulmonary nodules,5 miRNAs(mir-15b-5p,mir-17-5p,mir-19a-3p,mir-92a-3p,mir-146b-3p)were significantly overexpressed in peripheral circulation of non-small cell lung cancer patients.At the same time,5 miRNAs(mir-15b-5p,mir-17-5p,mir-19a-3p,mir-20a-5p,mir-146b-3p)were significantly overexpressed in peripheral circulation of non-small cell lung cancer patients compared to healthy population.2.While Logistic regression model indicated that age and miR-146b-3p were independent risk factors in the group of early non-small cell lung cancer and benign lung nodules.MiR-19a-3p and miR-146b-3p were independent risk factors in the group of early non-small cell lung cancer and healthy control.3.The diagnostic value of mir-146b-3p was evaluated by ROC curve,in the group of early non-small cell lung cancer and benign lung nodules: the area under the curve was 0.77.When the threshold value was 0.01,the sensitivity was 75% and the specificity was 71%.And the AUC of mir-panel was 0.81.When the threshold was 0.85,the sensitivity was 65% and the specificity was 89%.In the group of early non-small cell lung cancer and healthy control: the area under the curve was 0.93.When the threshold value was 0.72,the sensitivity was 89% and the specificity was 84%.And the AUC of mir-panel was 0.98.When the threshold was 0.90,the sensitivity was 94% and the specificity was 95%.ConclusionCompared with the benign pulmonary nodules and healthy group,mir-146b-3p was significantly overexpressed in peripheral circulation of non-small cell lung cancer patients.Mir-146b-3p is an independent risk factor for early non-small cell lung cancer,and it has certain significance for early non-small cell lung cancer,benign pulmonary nodules and healthy population,which can be used as a new blood marker in the clinic.
