| BackgroundAspirin(ASA),as a clinical commonly used NS AIDS drug,has antipyretic,analgesic and anti-inflammatory effects.In recent years,it has been found that aspirin(ASA)also has a role in inhibiting the development and metastasis of various tumors.In vitro studies have shown that Aspirin(ASA)can inhibit the expression of β-catenin protein in Wnt/β-catenin signaling pathway in human ovarian cancer and colorectal cancer,However it has little research on anti-tumor in combination with chemothera-peutic drugs,especially in anti-esophageal squamous cell carcinoma.ObjectiveTo investigate the inhibitory effect of aspirin(ASA)combined with cisplatin on proliferation of esophageal squamous cell carcinoma cell line Eca-109 and its related molecular mechanism,and to provide new theoretical basis for chemotherapeutic treatment of esophageal squamous cell carcinoma.Methods1.The Eca-109 cells were divided into blank control group(A),aspirin group(B),cisplatin group(C),aspirin + cisplatin group(D),group B(B-1 1.25 mmol/L,B-2 2.5 mmol/L,B-3 5 mmol/L,B-4 10 mmol/L,B-5 20 mmol/L),C group with cisplatin concentration 4.17μmol/L,D group with aspirin concentration 5 mmol/L.2.Using methyl thiazolyl tetrazolium(MTT)Methods detect cells proliferation inhibition rate in each group at different time(24 hours,48 hours,72 hours),and the interaction of the two drugs was calculated.3.The expression of β-catenin protein was detected by Western Blot after 48 hours by taking different treatment.Results1.MTT results showed that the inhibitory rate of each aspirin group on Eca-109 cells was significantly higher than that of the blank group at the same time point(P<0.05),and the proliferation inhibition rate of the Eca-109 cells was Concentration-dependent(P<0.05).At the same concentration,the proliferation inhibition rates were increasing with the time in different aspirin groups(P<0.05)and showed time-dependent(P<0.05).At the same time point,the inhibitory rate of aspirin combined with cisplatin was significantly higher than that of single drug aspirin(P<0.05)and single cisplatin(P<0.05).2.Western Blot showed that the expression of β-catenin protein in group B-3,group C and group D was significantly lower than that in group A after 48 hours of treatment,and the expression of β-catenin protein in group D was higher than that in group B-3,C group was significantly reduced(P<0.05).ConclusionsASA has an inhibitory effect on the proliferation of esophageal squamous cell carcinoma cell line Eca-109 in a time-dependent manner.ASA combined with cisplatin has a synergistic inhibitory effect on the proliferation of esophageal squamous cell carcinoma cell line ECA-109,and its mechanism may be related to the inhibition ofβ-catenin protein expression in Wnt/β-catenin signaling pathway... |
