Objective To investigate the effect of sodium glycididazoleradiosensitization on the efficacy of radiotherapy for rectal cancer and the mechanism.Methods The colorectal cancer cells SW480 were taken as the study subjects and were divided into the study group(treated with sodium glycididazoleand radiation),sodium glycididazole group(treated with sodium glycididazole),single group(treated with radiotherapy)and blank group(without any treatment).The cell growth inhibition and cell cycle were respectively detected with MTT and flow cytometry;ATMm RNA expression and ATM protein expression in each group were detected by immunoblotting hybridization.Results The 48 and 72 h cell growth inhibition rates in the study group and the single group were significantly higher than 24 h.The cell growthinhibition rates showed statistically significant differences at different time points(P<0.05).The cell growthinhibition rates in the study group at different time points were significantly higher than those in the single group and the sodium glycididazole group(P<0.05).The cell ratio in G0/G1 phase in the study group was significantly lower than that the sodium glycididazolegroup,the single groupandblank groupwhile the cell ratio in S phase was significantly higher than the sodium glycididazolegroup and the single group(P <0.05).The cell ratio in G2/M phase showed no significant differences among the four groups(P>0.05).The expression quantity of ATM m RNA and ATM proteinin study group was the lowest,the single group was the highest,and the expression quantity in the blank group and the sodium glycididazole group was no difference.Between the 4 groups,There was no statistical significance between the blank group and the sodium glycididazole group(P>0.05),In the remaining two groups of comparison,The difference of The other comparison of every two groups was statistically significant(P<0.05),Conclusion Sodium glycididazole is with strong sensitizing effect,and the mechanism is mainly related to the down-regulation of expression of SW480 cell ATM and cell cycle of regulation...
