Breast cancer is the most common type of malignant tumor in women.Metastasis is the main cause of breast cancer related death.Although great progress about the diagnosis and treatment of breast cancer has been made,the underlying molecular mechanisms of breast cancer metastasis is unclear.Recently,accumulating evidence has revealed that aberrant expression of lncRNA in different types of tumors,including breast cancer,was involved in occurrence and development of tumor.In this study,we analyzed of lncRNA expression in human breast cancer samples by Gene Expression Omnibus(GEO)database,and the expression of lncRNA FGF14-AS2 in 45 breast cancer tissues and their adjacent tissues was examined by qRT-PCR.The qRT-PCR indicated that compared with their adjacent tissues,the expression level of FGF14-AS2 was significantly downregulated in breast cancer tissues,and negatively correlated with clinical stages in patients.At present,the roles and molecular mechanisms of FGF14-AS2 in the occurrence and development of breast cancer has not been reported.We further analyzed the effect of FGF14-AS2 on the proliferation,migration,invasion of breast cancer cells by CCK8 assays,Wound healing assays and transwell assays.Effects of FGF14-AS2 on the metastasis of breast cancer was determined by tail vein injection using nude mice;QRT-PCR,Western blot assays and Luciferase reporter assays were performed to investigate the underlying mechanisms by which FGF14-AS2 regulates the expression of FGF14.The results showed that FGF14-AS2 has no abvious effects on the proliferation of breast cancer cells,but inhibited the migration and invasion of the cells.Furthermore,FGF14-AS2 inhibited breast cancer metastasis and functioned as ceRNA to regulate FGF14 expression by sponging miR-370-3p.In conclusion,it may be a biomarker for the prognosis of breast cancer patients.
