The Effects Of T4 And CL On Tafazzin And ALCAT1 In Hyperthyroidism Model

[Objective]Previous research of our group showed that the heart function is closely related to cardiolipin(CL)during the process of Hyperthyroid heart disease,but the detailed mechanism is not still unclear.As it is known,tafazzin and ALCAT1 are related to the reconstruction of CL,Therefore,this project aims to research the effects of T4 and CL on Tafazzin and ALCAT1 and get a further sight into our previous finding through the research about CL reconstruction.[Methods]The female KM mice were used in vivo and treated with T4 intraperitoneally for continuous 21 days to establish a hyperthyroidism experimental animal model.The changes of Tafazzin and ALCAT1 were detected by Real-time,Western blot and immunofluorescence.H9C2 cells were used in vitro to investigate the effects of T4 and CL on Tafazzin and ALCAT1.[Results]?In vivo experiments showed that T4 could increase the expression of Tafazzin both in protein and gene levels,and reduce the expression of ALCAT1 in protein level.? In vitro experiments showed that the expression of Tafazzin protein in H9C2 cells was initially increased by the treatment of CL and T4,then remained unchanged along with the time period;The expression of ALCAT1 protein was dreased,then increased along with the time period;the upregulation of Tafazzin caused by CL differed saturations,most significant upregulation in CL 14;0.? Dreased expression of Tafazzin protein and increased expression of ALCAT1 protein were caused after 6hs treatment of CL and T4 in cardiolipin synthase(CLS)knockdown H9C2 cells.[Conclusion]? T4 and CL can affect the expression of Tafazzin and ALCAT1 and this kind of effect can be influenced by CLS;? T4 shows consistant effect on Tafazzin with CL14:0:? The effects of both T4 and CL on the expression of Tafazzin and ALCATI are time related.
