Exosomes are cup-shaped or dish-shaped bodies membranous vesicles that are secreted outside the cell by intracellular multivesicular bodies that fuse with the cell membrane with diameter of 40 to 120 nm.Exosomes play an important role in immune monitoring neural remodeling tissue repair,stem cell survival,and coagulation function,contained various components such as lipids,proteins,and microRNA.Studies have shown that stem cell-derived exosomes can promote tumorigenesis and metastasis.However,some researchers have also put forward the opposite opinion that stem cell-derived exosomes inhibit the growth of tumors.Therefore,this paper focuses on the isolation and identification of exosomes derived from human iPS cells and the effect on proliferation,invasion and migration of human lung adenocarcinoma A549 cells.Research objective:Isolation of exosomes derived from human iPS cells and the effect on proliferation,invasion and migration of human lung adenocarcinoma A549 cells.Research methods:Improve culture conditions of human iPS cells and human lung adenocarcinoma A549 cells.Exosomes derived from human iPS cells were isolated by ultracentrifugation from the supernatant of human iPS cells,and the morphology of the exosomes was observed by transmission electron microscopy.The expression of transmembrane proteins on the surface was detected by Western blot.The effects of exosomes on the proliferation and invasion of human lung adenocarcinoma A549 cells were detected by MTS method and Transwell method.The effect of exosomes derived from human iPS cells on the migration of human lung adenocarcinoma A549 cells was determined by cell scarification.Research resultsand conclusion:(1)Exosomes derived from human iPS cells were isolated from the supernatant of human iPS cells.Electron microscopic observation of exosome is round or oval cup shape,uniform size,diameter of about 100nm.Western Blot identification of the exosome surface is rich in protein CD63;The results of MTS showed that exosomes derived from human iPS cells promoted the proliferation of A549 cells and were time-and concentration-dependent(P<0.05).Transwell method results showed that exosomes derived from human iPS cells had the effect of enhancing the invasion ability of A549 cells(P<0.05);Cell scarification test results showed that exosomes derived from human iPS cells had the effect of promoting migration of A549 cells(P<0.05).(2)The results showed that exosomes derived from human iPS cells can promote the proliferation of human lung adenocarcinoma A549 cells,enhance their invasion and migration ability.
