Background Calcific aortic valve disease(CAVD)has become the third most common cardiovascular disease in developed countries.There is no specific drug treatment.Surgery and interventional therapy are the most effective treatments.Advanced glycation end products(AGEs)is Mailland reaction(non enzymatic glycosylation reaction),the product of western countries on a high-fat diet way of life can cause a lot of glycosylated end products produced in the body.AGEs can cause a variety of pathological processes,including atherosclerosis.It is reported that atherosclerosis and calcified aortic valve disease have similar pathological processes and risk factors,but there is no evidence that AGEs have a clear correlation with CAVD.Hydroxyl methyl lysine(CML,which kind of,representative)in the valve deposits form similar to atherosclerosis of the aorta,which receptor(RAGE)in which mediatedsignaling,inflammatory response and a series of play an important role in the process.The other team members for animal experiments have established that a high-fat diet causes of ApoE-/-mice in the aortic valve which increases,mediated by RAGE signal makes the mass transformation between the early emergence in the aortic valve endothelial(EndMT),eventually lead to appear the aortic valve calcification.Based on the above academic foundation,therefore,this topic main research objective is to observe the CML and its receptor expression levels and calcification the relationship between different degree of pathological changes in patients with aortic valve disease compared with non calcified CML expression level in patients with aortic valve disease to explore which influence on aortic valve calcification sex disease,for further study of aortic valve calcification(aortic valve calcification,AVC)pathogenesis provides a theoretical basis for clinical.Methods Screening qualified calcification of aortic valve disease,and conform to the conditions of the aortic valve lesions of patients,divided into normal control group(N group),light calcium group(group S),moderate calcification group(M),severe calcification group(group H),serum samples from patients and the pathological changes of aortic valve replacement after valvular specimens;To analyze the relationship between blood glucose and blood lipid,valvular ultrasound integral,valve tissue protein imprinting and immunofluorescence staining and the degree of calcification of aortic valve.Results There was no significant difference in blood glucose and blood lipid in each group.In this study,the relationship between blood glucose,blood lipid level and AVC level was not statistically significant.There was significant difference in the cross-aortic valve velocity and cross-valve pressure difference between the patients in each group,and the change trend was consistent,indicating that the degree of calcification was more severe,and the higher the cross-lobe pressure difference and the cross-lobe flow rate.The accumulation of CML in the aortic valve in patients with mild-to-moderate mainly appear on the edge of the valve,in patients with moderately severe has spread to valve inside,but in some patients with severe calcification,the accumulation of CML fell;The protein expression levels of RAGE in each group gradually increased with the increase of calcification degree,but the moderate calcification group increased the most,and some severe calcification groups decreased.Groups associated with calcification in the ALP expression level rise,according to the degree of calcification of but in severe calcification group,ALP expression level decreased,ALP and alpha SMA in fluorescent results orientation,are first appeared on the edge of the valve,then gradually extends to internal;Associated with mass transformation between endothelial protein VE-cadherin and alpha protein imprinting and immunofluorescence results trend of SMA,between groups all have different degrees of EndMT,VE-cadherin expression level with prolonged gradually reduce,the alpha of the SMA expression level increased gradually,that EndMT increased with calcification degree aggravating,but no significantly statistical differences.However,there was a decrease in the protein expression in patients with severe calcification group,and the reason for this was still to be further explored.Conclusion Expression level with the progress of the degree of calcification,which is on the rise,the expression of RAGE quantity also gradually increase,but in the patients with severe calcification,which and the expression of RAGE amount is rather low,with fibroblasts into osteogenesis mesenchymal cells,and led to the decrease of the amount of protein expression calcium salt deposits...
