Objective: Determination of different stages of distribution and expression of P53 protein in patients with acute leukemia,and explore the occurrence,development and outcome of acute leukemia,and study the relation to response to conventional chemotherapy and clinical parameters related to prognosis,provide the basis for the assessment of the prognosis of leukemia,and provide a new target for gene therapy of leukemia.Method: The collection of 50 cases of acute leukemia(AL)patients,30 patients of initially treated was set initial treatment group,20 patients with recurrence was set recurrence group and 30 cases of bone marrow in normal healthy subjects as control group.The collected cases were all diagnosed by clinical and related auxiliary examinations and met the WHO diagnostic criteria for leukemia.By using the method of enzyme linked of Streptavidin perosidase(SP)determination of P53 protein expression of bone marrow cells in each group,comparing the expression of P53 protein positive rate between groups.Patients with positive expression P53 were assigned to the P53(+)group,patients with negative P53 negative were assigned to the P53(-)group,with 50 cases of AL patients one standard chemotherapy induced remission,and the curative effect evaluation.Among them,comparing P53(+)group and P53(-)group remission rate.Using SPSS 17.0 statistical software,to analyze the data processing,count data through the cases and the percentage(%),said the ?2 test was used to the comparison between the two groups,with P < 0.05 showed significant difference,statistically significant.Results:1.50 patients with AL,P53(+)has 15 cases,the total expression rate of 58%(29/50),significantly higher than that of healthy group: 0%(0/30),the difference was statistically significant(P<0.05);and the recurrence group positive expression rate higher than those in initial treatment group(P=0.047);2.Among 50 AL treated patients,27 cases of patients achieved CR,the total CR rate was 54%(27/50),comparing before chemotherapy,the positive expression rate of P53 is decreased after chemotherapy(P=0.001),the remission rate of P53(-)group was obviously higher than that of P53(+)group(P=0.034);3.The expression of P53 was related to the patient's white blood cell count,hemoglobin,platelet count,and whether there was extramedullary infiltration in AL patients,but not sex.Conclusions: The expression of P53 protein in patients with AL positive rate increased significantly,and is closely related to the clinical curative effect after chemotherapy and prognosis,the clinical effect of chemotherapy patients according to the evaluation of the level of expression of P53,help us to judge the prognosis of the disease,with P53 as the target to become the new targeted therapy by means of drug substitution and recovery P53 the function,so as to improve the prognosis of patients.
