Objective:To study the effects of compressive force on GSK-3?/?-catenin signal shaft and the expression of ?-SMA in human gingival fibroblasts(HGFs)cultured on PLGA Scaffold,in order to explore the possible mechanism of the mechanical signal transduction of HGFs cultured on PLGA Scaffold.Methods: 1.The HGFs was cultured,the PLGA-HGFs was constructed and loaded with 25g/cm2 compressive force.According to the time of adding force,it was divided into 0h(control group),6h,24 h,48h,72 h group.The m RNA and protein expression levels of GSK-3?,?-catenin,?-SMA were tested by RT-q PCR and Western blotting methods,respectively.2.After subjected Li CL and compressive force(25g/cm2).The m RNA and protein expression of GSK-3?,?-catenin,?-SMA were investigated by RT-q PCR and Western blotting methods.Results:1.The results after loading force:(1)GSK-3? expression level showed a general decline trend,except m RNA 72 h group and protein 48 h group,compared with the control group,the difference was statistically significant(P < 0.05);(2)The expression level of ?-catenin was up rapidly,the 6h group reached the peak value,except the m RNA 72 h group,the difference was statistically significant(P<0.05)compared with the control group(P < 0.05);(3)The expression of m RNA of?-SMA in a increased and increased significantly in 72 h group.The expression of protein of ?-SMA was down,and then continued to decline after a small increase.There was a significant difference between the control group(P < 0.05).2.After adding Li CL and force,the results showed:(1)The expression level of GSK-3?m RNA was higher than that of non inhibition group before group 24 h,and in 48 h group was lower than that in non inhibition group.The protein expression level was higher before 48 h,and the 72 h group was lower than that of non inhibition group.Except m RNA 48 h,72h group and protein 48 h group,there was significant difference between groups(P < 0.05).(2)The expression of ?-catenin m RNA and protein increased rapidly and the 6h group reached the peak value.Except for the m RNA 72 h group,the expression level of m RNA and protein in the inhibition group was higher than that in the non inhibition group,and the difference between the groups was statistically significant(P < 0.05);(3)The expression of?-SMA was down,the 72 h group m RNA overexpressed but lower than the non inhibition group;the inhibitor protein expression was generally lower than that of the non inhibitory group,except for m RNA 48 h group and protein 0h and 24 h group,the difference was statistically significant(P < 0.05).Conclusions: Compressive force can be transmitted by GSK-3?/ ?-catenin signal shaft of HGFs and affect the expression of ?-SMA.GSK-3 ?/ ?-catenin signal shaft was correlated with the expression of ?-SMA.The expression of?-SMA is positively correlated with the expression of GSK-3? in the early addition of force and is negatively correlated with the late addition of force,and has a continuous negative correlation with the expression of ?-catenin.
