Objective To screen differentially expressed RNAs in asopharyngeal carcinoma CNE2 cells induced by hyperthermia and normal CNE2 cells by high-throughput RNA sequencing,and to provide a theoretical basis and genomic strategy for molecular mechanism,targeting therapy and prognosis prediction for nasopharyngeal carcinoma.Methods 1.The cells were divided into two groups: normal group and hyperthermia group.Cells in hyperthermia group were subjected to water-bath hyperthermia at a temperature of 42? for a period of 60 min.immediately following hyperthermic treatment,the cells were equilibrated at 37?.2.Total RNA of two groups were extracted and the qualities of two RNA samples were detected.3.The Illumine HiSeqTM2500 sequencing platform was used for high-throughput transcriptome sequencing.Then,the original date was sorted,filtered and quality evaluated.4.IPA analysis were used to analyze the transcriptome sequencing results to identify differentially expressed RNAs in asopharyngeal arcinoma CNE2 cells induced by hyperthermia and normal CNE2 cells.Functional enrichment analysis of differentially expressed genes wereperformed by GO(Gene Ontology)and KEGG(Kyoto Encyclopedia of Genes and Genomes)Results In this study,3184 differentially expressed genes were identified through high-thoughput transcriptome sequencing technology,of which 2712 genes were up-regulated and 472 genes were down-regulated,and 90 signaling pathways,25 Gene-Coexpression-Network of IPA and 466 upstream regulator were closely related with tumorgenesis and development,which associated with molecular function,cell component and biological process.Gene GO and KEGG enrichment analysis showed that differentially expressed genes were significantly involved in the biological processes of extracellular matrix organization,cell growth,extracellular structure organization,locomotion,extracellular matrix disassembly,as well as a number of cancer-related signaling pathways.Conclusion There are 20 differential genes(MMP3,BCAT1,CXCL12,FLT1,KCNMA1,MME,GREM1,ect.),5 upstream regulator(TGFB1,TNF-?)and 7 signaling ways(NF-?B signaling pathway,PI3K-Akt signaling pathway and MAPK signaling pathway,ect.)were found in this study to be correlated with the tumorigenesis of asopharyngeal carcinoma,suggesting these differentially expressed genes may be targeting therapy of asopharyngeal carcinoma and provides new ideas for the clinical treatment of nasopharyngeal carcinoma.
