Objective:The rare-earth magnesium alloy(Mg-Nd-Zn-Zr)has a good application prospect as a neurosurgical material due to its biodegradability and excellent mechanical properties.This topic mainly studies the biocompatibility of rare earth magnesium alloy on nerve cells.Method:1.Preparation of rare earth magnesium alloy extracts with different dilution concentrations;2.Primary culture and purification of astrocytes and neural stem cells,and identification by immunofluorescence;3.The effects of rare earth magnesium alloy extracts on the activity of astrocytes were evaluated by MTT method.The cell cycle of astrocytes was detected by flow cytometry,and the expression of related proliferative proteins was detected by Western blot..4.The neural stem cells were tested for the cytotoxicity of the rare earth magnesium alloy extracts after one day and three days by cck-8 method.The Q-PCR assay extracts of each group on the neural stem cells PCNA,BAX,BCL-2 In gene expression,neural stem cells were finally induced to differentiate at the concentration that promoted neural stem cell differentiation,and differentiated cells were identified by immunofluorescence.result:1.The results of cytotoxicity showed that rare earth alloy extracts showed no toxic effects on both astrocytes and neural stem cells.Rare earth magnesium alloy extracts have good biocompatibility with diluted cells at dilution ratios of 3 x and 10x.2.The results of cell proliferation experiments showed that the rare earth magnesium alloy extract diluted at low concentration(10x)promoted the proliferation of astrocytes and neural stem cells.When cultured at high concentration(1x),astrocytes and The growth of neural stem cells was slightly inhibited compared with the negative control group.The experimental results of neural stem cells differentiation induced by rare earth magnesium alloy extracts showed that dilute 10x group of rare earth magnesium alloy extracts could promote the differentiation of neural stem cells into astrocytes and neurons.in conclusion:Rare earth magnesium alloy extract can promote the proliferation of astrocytes at the dilution of 10x and PCNA protein expression at the 3x group.Rare earth magnesium alloy extract can promote the proliferation of neural stem cells and the upregulation of PCNA gene expression when diluted 10x.At the same time,Bax and bcl-2 genes were down-regulated.However,in the 1x and 2x groups,astrocyte cell cycle and neural stem proliferation were affected by high magnesium alloy concentrations.According to the experimental results,it can be seen that Mg-Nd-Zn-Zr alloy has good biocompatibility with nerve cells and can be used for the next animal experiment.
