Objective: To investigate the effects of ABVD,a first-line chemotherapy drug for Hodgkin's lymphoma,on reproductive system and reproductive function in female sexually mature mice, and provide guidance for the clinical application of ABVD.Methods: A saline control group(NC group)and an ABVD combined chemotherapy group(ABVD chemotherapy group)were established.ABVD combined with chemotherapeutic drugs was established by intraperitoneal injection of ABVD.The model of ABVD combined with chemotherapy was established. The saline control group was injected intraperitoneally with the same dose of physiological saline.Serum levels of AMH,uterine weight, and ovary weight were measured, and morphological features of the ovaries,as well as changes in the number of follicles at each level, were observed. After 4 weeks of cessation of administration, observing changes in the estrous cycle of the two groups of mice, detecting the AMH level in the mouse serum, and the changes of the uterus weight and ovary weight, observing the morphological characteristics of the ovary, and the number of follicles at all levels.change.Intraperitoneal injection of pro-drugs for pro-drug stimulation,in vitro fertilization(IVF)or cage closure was performed. The recovery of fertility in mice was examined by observing the success of the mice's conception and the number and survival rate of offspring. Results: After administration, the estrous cycle of ABVD combined with chemotherapy group was disordered and mostly stagnated during the intercourse. After four weeks of administration, weight gain was significantly lower in the control group(P<0.01),serum AMH levels were significantly lower than in the control group(P<0.01),and ovarian mass and uterine mass were lower than in the control group(P<0.01).The proportion of mature follicles in Central Africa increased, compared with the control group, there was a significant difference(P <0.01).After 4 weeks of cessation of administration, weight gain, uterine mass, ovarian mass, and estrus cycle of the mice were restored. The results were not statistically different from those of the control group. After 4 weeks of drug withdrawal, serum AMH levels in mice increased, which was significantly different from that in the control group(P<0.01).After discontinuation of treatment, the number of offspring of offspring in the ABVD combined chemotherapy group was less than that of the saline control group(P<0.01);however,there was no significant difference in the birth weight of the offspring between the two groups. The number of follicles at each level in the ABVD combined chemotherapy group was similar to that of the normal group. Conclusions: After ABVD combined with chemotherapy, the estrus cycle is disordered, the ovarian function is significantly declining, the ovarian tissue and AMH levels are significantly reduced, and it has an inhibitory effect on the reproductive system of mice, which is similar to that of patients with clinically advanced ovarian failure.Four weeks after the ABVD chemotherapy was stopped, the ovarian function of the mice was restored, the AMH level was elevated, and the number of follicles at all levels returned to normal levels.ABVD combined with chemotherapy can significantly inhibit ovarian function in female mice. After cessation of chemotherapy, both ovarian function and reproductive function can be restored in mice.
