Objective:Make SD rats inhaled with different drugs,observe their lung pathological change through light microscopy,to evaluate the safety of different drugs inhaling on natural rats and provide laboratory evidence for clinical mdication.Methods:40 rats were randomly divided to eight groups,every group had 5 rats.Which were separate inhaled with A:nothing?B:Normal Saline Solution(10mL normal saline solution)?C:Salbutamol Sulfate Nebules Inhalation Solution(5mg/20 mL salbutamol aulfate nebules inhalation solution2mL+8mL normal saline solution)?D:Dingchuantang(10mL polyjuice,whose concentration was 10g/mL)?E:Shuanghuanglian Power-injection(10mL potion,whose concentration was 20mg/mL)?F:Gentamicin(8U/2mL gentamicin4mL+6mL normal saline solution)?G:Compound Salvia Injection(10mL compound salvia injection)?H:Silicon Dioxide(10mL potion,whose concentration was 40mg/mL),twice a day,last 56 days totally.Then,blood and bronchoalveolar lavage fluid were collected and analyzed for cell count,percent of each type of cell,to measure the severity of the inflammatory.Additionally,Histopathology revealed the lung's pathological change and the number of dust cell;immunohistochemistry revealed CD163 responding.Results:(1)White blood cell count:A(3.96±0.36)×10~9/L,B(4.66±0.58)×10~9/L,C(4.06±0.86)×10~9/L,D(8.98±1.08)×10~9/L,E(7.10±0.88)×10~9/L,F(6.14±0.89)×10~9/L,G(9.84±2.33)×10~9/L,H(8.99±2.48)×10~9/L,comparative analysis between the 8 group had significant difference(P with statistical significance);differences among A?B and C were not significant;white cell count in BALF:A(2.16±1.04)×10~9/L,B(3.94±0.67)×10~9/L,C(4.36±1.15)×10~9/L,D(14.58±2.93)×10~9/L,E(19.68±6.29)×10~9/L,F(11.74±1.03)×10~9/L,G(44.75±10.8)×10~9/L,H(53.54±14.25)×10~9/L,comparative analysis between the 8 group had significant difference(P with statistical significance);differences among A?B and C were not significant;lymphocyte count in BALF:A(18.70±9.00)×10~8/L,B(36.01±5.99)×10~8/L,C(38.95±11.69)×10~8/L,D(132.70±26.94)×10~8/L,E(173.56±57.6)×10~8/L,F(106.60±16.76)×10~8/L,G(340.63±70.97)×10~8/L,H(495.63±131.95)×10~8/L,comparative analysis between the 8 group had significant difference(P with statistical significance);differences among A?B and C were not significant;(2)number of lung dust cell count in 10 sight of high light microscopy:A 12?B 26?C 17?D 262?E 133?F 109?G 96?H 315,comparative analysis between the 8 group had significant difference(P with statistical significance);differences among A?B and C were not significant;(3)hematoxylin-eosin staining of lung:A?B and C had no pathological change in the lung,but C?D?E?F?G and H had pathological changes of different degrees;(4)immunohistochemistryofCD163responding:A?B and C were negative expression,C?D?E?F?G and H were positive expression of different degrees.Conclusions:The rats inhaled with Normal Saline Solution and Salbutamol Sulfate Nebules Inhalation Solution had no pathological changes in lung,as well as they had a same degree expression of cell count in blood and BALF,which showed that inhaling with Normal Saline Solution and Salbutamol Sulfate Nebules Inhalation Solution were safe;however the other drugs could lead to different degrees pathological changes in lung and high expression of cell count in blood and BALF,which read inhaling with the drugs which were not aerosol formulations were not fafe.
