Objective:To explore the selection of eligible donors for fecal microbiota transplantation(FMT).After screening,exploring the effective preservation of fecal microbiota products,and providing the basis for further study on the preservation and transportation of the fecal microbiota products.Methods:FMT donors were selected according to a three-step approach-recruitment,screening and medical.examinations.The fresh fecal microbiota products were divided into 6 groups according to the different methods of adding glycerol and sucrose protective agent or making freeze-dried powder,diluted the fecal microbiota product with decimal dilution method,apllied each concentration of fecal microbiota product to Wilkins-Chalgren medium with a simple drop plate method,cultured in the30?thermostat and counted.The microbiota products were resuscitated for 2 weeks,1 month,2 months,and 3 months,and cultured and counted in the same way.Results:A total of 131 volunteers were recruited in the study.102 volunteers were excluded after viewing their questionnaire responses and the pass rate was 22.14%.According to medical examination results,5 volunteers were eligible for FMT and the pass rate was 17.24%.Based on our research,the final pass rate for FMT donor selection was 3.82%(5/131).The number of viable cells in the fecal microbiota product decreased with time,and the descending rate was the fastest in the first two weeks,decreasing by 105-107.Conclusions:Screening for qualified donor is a difficult task with high cost and low pass rate.The study provided a practical method for further selection of FMT donors.There is little differens in the preservation methods,including protective agent of glycerol,sucrose,and lyophilized powders.This study also provides the basis and practical method for the preservation and transportation of fecal microbiotal products.
