Objective:The purpose of this experiment is to explore and study the intervention mechanism of Pingchuanning to the cold asthmatic rats airway remodeling,which effected to related proteins in the PI3K-Akt/PKB signal transduction pathway.By immunohistochemistry and Rt-PCR methods to observing the proteins PIP3,PDK2,IKK?,p110?,CDK2 expression level in cold asthmatic rats lung tissue and the intervention of Pingchuanning on its expression level,at the same time to explore the mechanism of Pingchuanning to inhibit the cold asthmatic rats airway remodeling.Method:140 SPF and healthy male SD rats(6-8 weeks,weight 180-220 grams)were selected to random grouping and divided into 7 groups,model group,normal group,dexamethasone group,Guilongkechuanning group,Pingchuanning high-dose group,Pingchuanning middle-dose group,Pingchuanning small-dose group,each group of an average of 20 rats.In the 1d,8d after the end of adaptability raise,separately to every rat(the normal group rats injected 1ml saline into the abdominal cavity of the rat)by intraperitoneal injection 1ml configured mixed suspension to make them sensitization,the mixed suspension by physiological saline,aluminum hydroxide,and10%OVA.From the 15 d,1%OVA solution was used for the ultrasonic atomization of the rats continuing 1 week,and the rats were exposed to cold stimulation for 30 min each day.Since the 21 d,the rats were given gastric medicine treatment,continuing28 d,five treatment group rats to filling dosage,conversion volume based on rats and human body surface area,the normal group and model group rats daily irrigation served the same amount of normal saline.After 28 d lavage dosing,line with OVA for the last time excitation and within 2h dissect rats,the right lung tissue of rats stored with-80?,formalin fixed left lung tissue to be detected.Results:With HE staining picture,we find the lung tissues of rats in the model group showed inflammatory cell infiltration,thickening of airway mucous folds and narrowing of airway,which were more obvious than normal group.The other five treatment groups were inhibited by the above phenomena,compared with the model group.Immunohistochemical detection:compared with normal group,the expression level of p110? and CDK2 in the remaining 6 groups was significantly increased,with statistically significant(P<0.05,P<0.01).Compared with the model group,the expression level of p110? and CDK2 in the lung tissues of the five treatment groups decreased(P<0.05,P<0.01).Compared with the three groups in the large,medium and small dose group,the treatment effect of the large and medium dose group was better than that of the low-dose group(P<0.05,P<0.01),especially the large dose group of lung tissue p110?,CDK2 to cut the most prominent expression level,and compared with the effect of dexamethasone group,Guilongkechuanning group the better(P<0.05,P<0.01),the more significant statistical significance.Rt-PCR method was used to detect the expression levels of PIP3,PDK2 and IKK? in the other six groups(P<0.05,P<0.01)compared with the normal group.Compared with the model group,the expression levels of PIP3,PDK2 and IKK? in the five treatment groups were down-regulated(P<0.05),and the reduction was most significant in the large dose group.Conclusion:The expression of PI3K-Akt/PKB pathway p110?,CDK2 can be controlled by Pingchuanning,and the expression level of PIP3,PDK2 and IKK? were down-regulated by Pingchuanning.Thereby,reducing the airway inflammation response,inhibiting the bronchial hyperresponsiveness and slowing the airway reshape process to treat cold-asthma.
