Objective: To study the contrast of Juantong Decoction,Juan-carousing decoction in treating endometriosis(EMs)model rats with Juantong Decoction with LY294002,and to observe the gene expression of the related factors in the cell PI3K/AKT/mTOR signaling pathway by Juan Carousing Decoction,At the same time,the effect of 4EBP1 protein expression was observed to elucidate the therapeutic effect of the decoction on the endometriosis,in order to provide a new idea for elucidating the pathogenesis of endometriosis,and to provide a theoretical basis for TCM treatment of endometriosis.Methods: The model of endometriosis SD rat was established by the method of autologous endometrial transplantation,and the effects of different dosage of water decoction and Chinese herbal blocker on the model rats were observed by using the model.Female rats with normal estrous cycle were selected as subjects,randomly divided into 6 groups,8 in each group,the normal sham operation group,named of the Model control group,the Juantong high dosage group,the Juantong medium dosage Group,and the Juantong low dosage group and the Channel blocker(LY294002)group..Results:1 The results of RT-PCR were used to detect the PI3K,Akt,MTOR and4ebp1 mRNA in rats,and compared with normal group,the relative expression of PI3K mRNA in each group was obviously elevated,there was significant difference and statistically significant(p<0.05).Compared withmodel group,The expression of low dose Group,medium dosage Group,high dosage Group,the Channel blocker Group had significant difference(p<0.05),and the effect of the low dosage group,the middle dosage group and the high dosage group was significantly different from that in the Channel blocker Group(p<0.05).2 The expression of 4EBP1 protein content in rat endometriumThe results of Immunohistochemistry showed that the expression of4E-BP1 protein was significantly higher in all groups of rats than in normal group,and had significant difference with model group and Low-dose Group,which was statistically significant(p<0.05).There was no significant difference between the normal group and the middle Dosage group,the high dose group of Juantong Decoction and the Channel blocker Group(p>0.05).Compared with the model group,the expression of low dosage,medium dosage,high dose,the Channel blocker Group was significantly different(p<0.05).There was no significant difference(p>0.05)between high dose Group and medium dosage group,but there was significant difference(p<0.05)with the Channel blocker Group.There was no significant difference between high dose group,middle dose group and the Channel blocker Group(p>0.05).Conclusions:1 The gene expression of PI3K,Akt,MTOR and 4EBP1 mRNA in the rat ectopic endometrium was reduced by the way of Juantong;2 Juantong can reduce the expression of 4EBP1 protein in ectopic endometrium of rat model rats;3 Juantong may be possible to play a role in the treatment of endometriosis model rats by inhibiting the activation of pi3k/akt/mtor signaling pathways.
