Objective:To assess the expression of DPP8 in cervical cancer,corresponding normal cervical samples and cervical cancer cells,and the relationship between DPP8 and clinicopathologic features of cervical cancer.To investigate the effects of DPP8 on the ability of proliferation,apoptosis,invasion and mobility of cervical cancer cells as well as the possible mechanisms.Methods:To study the role of DPP8 in the initiation and progression of cervical cancer,we examined DPP8 levels in cervical cancer tissues and cells.Subsequently,cervical cancer SiHa and HeLa cells were treated with siRNA-DPP8 or pcDNA3.1-Flag-DPP8.We used cell cycle assay,5-Ethyl-2'-deoxyuridine proliferation assay,cell apoptosis assay to determine the effect of DPP8 on the proliferation and apoptosis of cervical cancer cells.To determine whether DPP8 regulates invasion and migration of cervical cancer cells,we used transwell assay to assess the number of cancer cells through matrix,real-time PCR and Western blot analysis to analysis the expression of their related proteins to determine the phenotype caused by depletion or overexpression of DPP8 in cervical cancer cell.Results:DPP8 expressions were upreg?lated in cervical cancer tissues compared to normal cervical tissues.Overexpression of DPP8 protein related with tumor differentiation,FIGO stage and lymph node metastasis for cervical cancer patients.DPP8 expression was observed in the cytosol and in the perinuclear area,as well as in the nuclei of cervical cancer cells.Notably,when cells were treated with siRNA-DPP8,the expression of BAX increased,and the expression of Cyclin Dl,Bcl-2,MMP2 and MMP9 was downregulated.In cervical cancer cell lines,silencing the expression of DPP8 not only suppressed the proliferation,migration and invasion of the cervical cancer cells,but also promoted cervical cancer cell apoptosis.When expression of DPP8 was up-reg?Lated,the opposite phenomenon can be observed.Conclusion:Analysis of series of evidences showed that DPP8 may be a usef?L and promising biomarker for patients with cervical cancer.
