Objective:To investigate the modeling method of rat model of type 2 diabetes;type 2 diabetes mellitus and discuss the intestinal tissue sodium glucose cotransporter 1(SGLT1)the relationship between the expression of contrast;acarbose(glucobay)and regulation of glimepiride in type 2 diabetic rats blood glucose levels,and the expression of SGLT1 in the jejunum tissue of rats in the discussion of the relationship between the expression of SGLT1 protein;acarbose and jejunum tissue of type 2 diabetic rat model.Methods:40 SPF adult male SD rats with normal feeding after a week,take a random number method is divided into five groups:normal control group,normal control +acarbose intervention group,model control group,type 2 diabetes mellitus type 2 diabetes mellitus model + BAL Tang Ping intervention group,type 2 diabetes mellitus model + glimepiride intervention group 8 rats in each group.Type 2 diabetes mellitus model control group and type 2 diabetes mellitus drug intervention group were treated with high sugar and drinking water combined with high fat emulsion continuously for 4 weeks to induce insulin resistance in rats.Normal group was given normal drinking water and equal dose of normal saline.Then type 2 diabetes mellitus model control group and type 2 diabetes mellitus drug intervention group were given abdominal abdominal injection of small dose of streptozotocin(STZ)for type 2 diabetes rat model establishment.After three days,the random blood sugar of rats was measured,and the value was more than 16.7mmol/l,then the rats of type 2 diabetic rats were successfully established.After the success of modeling,the intervention group rats were drug acarbose and glimepiride continuous drugs by gavage for 4 weeks intervention;then give equal dose of saline control group.The basic situation of rats was observed at any time,and the weight and blood sugar level of the rats were measured regularly.After the intervention was completed,the rats were killed and blood samples from the blood were collected by the heart extraction method.The centrifuge was used to separate the serum and keep the reserve.At the same time,the intestinal tissues of the rats were retained and one of the 4%polycondensation Formaldehyde Solution and liquid nitrogen was preserved for the later experiment.Finally,the expression of SGLT1 protein in jejunal tissues of each group was detected by Western blot method.The expression level of SGLT1mRNA in jejunal tissues was detected by qRT-PCR and immunohistochemical methods.Results:1、Compared with normal control rats,the blood glucose of type 2 diabetic model control group increased significantly by 78%,and significantly increased the expression of SGLT1 gene and protein in jejunal tissues,the difference was statistically significant(P<0.05).2.After 4 weeks of drug intervention,compared with the model of type 2 diabetic rats in the control group,the model of type 2 diabetes mellitus + acarbose intervention group,type 2 diabetes intervention model + glimepiride group could reduce blood glucose in type 2 diabetic rats,glucoby intervention group decreased 46%,glimepiride intervention group decreased 24%;type 2 diabetes mellitus model +acarbose intervention group,type 2 diabetes model + glimepiride group were able to express SGLT1 jejunal tissue of rats with down-regulation of gene and protein(P<0.05),at the same level of blood glucose,compared the intervention effect of Tang Ping worship and glimepiride that worship Tang Ping had more significant effect on the expression of SGLT1 protein in intestinal tissue of type 2 in diabetic rats.Conclusions:1.The model of type 2 diabetic rats can be successfully established by feeding SD rats with high fat emulsion and high sugar drinking water and injecting small dose of STZ into the abdominal cavity.Compared with the normal control group,the blood sugar of the rats in the model control group of type 2 diabetes was significantly increased.2.The expression level of SGLT1 gene and protein in the jejunum tissue of model 2 diabetes model rats increased significantly.The acceleration of intestinal glucose absorption mediated by SGLT1 may be one of the reasons for the development of patients with type 2 diabetes.3.Acarbose,glimepiride treatment effect of drugs to reduce blood glucose of type 2 diabetes rats and has certain effect on the expression of SGLT1 protein in the jejunum tissue of type 2 diabetic rats in the two groups can lower the expression of SGLT1 in type 2 diabetic rats jejunum tissue gene and protein.At the same level of blood glucose,compared the intervention effect of Ping Tang worship and glimepiride drugs showed that acarbose had more significant effect on the expression of SGLT1 protein in the jejunum tissue of type 2 diabetic rats model,suggesting that acarbose in type 2 diabetes rat model of intestinal tissue in a relationship between the expression of SGLT1 protein.
