Object:To investigate the therapeutic effect and mechanisms of novel compound Oleoylethanolamide(OEA)on type 2 diabetes(T2DM).Method:70 male Kunming mice were randomly divided into normal group(n=10)and T2DM group(n=60)after adaptive feeding for one week.The normal group treated with regular diet.however,the T2DM model group was established with high fat diet(HFD)and a low dose of streptozotocin(STZ).After T2DM model was built the model mice were divided into five groups(n=10 for each group):model,glipizide,OEA(low,medium and high does)group.They were received an intraperitoneal injection of Vehicle,glipizide(2.5 mg?kg-1?d-1)or different concentration of OEA(5,10 or 20 mg?kg-1?d-1)for 6 consecutive weeks.The level of fasting blood glucose(FBG),insulin(FIns)and insulin resistance index(HOMA-IR)were measured in the course of the experiment.Serum total cholesterol(TC),triglyceride(TG),free fatty acid(FFA)and superoxide dismutase(SOD)were determent and the expression of Glut4,TNFa,CREB,Erk and NF-KB in liver were analyzed by Western blot at the end of treatment.Result:Compared with the normal groups,the level of FBG,Fins,HOMA-IR,serum TC and TG obviously increased in model groups.The level of the expression of Glut4,TNFa,CREB,Erk and NF-kB in liver were obviously increased in model groups too.However,these results were reversed by OEA treatment.(P<0.01).By the way,these effects were similar to glipizde(P>0.05).Conclutlion:OEA ameliorated insulin resistance in a manner associated with increased intrahepatic glucose transport and inhibited inflammatory response.
