The Correlation Of MTHFR Gene Polymorphism With Primary Angle Closure Glaucoma

Objective This research aims at the determination of MTHFR in Guangxi region polymorphism of PACG gene C677T,to investigate its relationship with PACG.This is further to accumulated research data for the pathogenesis and early diagnosis of PACG,and as a specific biological indicators for screening and protection of susceptible population.Methods We collected 60 cases Patient of PACG hospitalized and 60 cases of normal controls in physical examination center from December 2013 to December 2014 in the Guangxi Zhuang Autonomous Region people's Hospital.Fasting venous blood was extract from the patients.And then extracted DNA from the blood.The MTHFR gene C677T were analyzed by the PCR-based restriction fragment length polymorphism(PCR-RFLP)method.Finally,the difference of MTHFR gene polymorphism between PACG and the normal controls were analyed by statistical software SPSS 19.0.Hardy-Weinberg analysis was performed by comparing the observed and expected genotype frequencies in study groups using Chi-square test for Whether the sample is representative of the population.Comparison of the MTHFR C677T allelotype distribution in the study groups was performed by means of two-sided contingency tables using Chi-square test.The level with P<0.05 was statistically significant.Results 60 patients and 60 normal controls are included in the analysis.The frequency distribution of two genotypes(CC,CT,TT)respectively are 88.3%?5%.6.7%and 98.3%.1.7%.0,of which the differences were statistically significant(P<0.05);the distribution frequency of TT genotype of MTHFR gene in the case group is significantly higher than that of the control group(P<0.05);the frequencies of T and C alleles of MTHFR gene are 90.8%and 9.2%,respectively;while for the control group,they are 99.2%and 0.8%.Obviously,there is difference between the two groups(?2=8.8,P<0.05).Conclusions The distribution frequency of TT genotype of MTHFR gene in PACG patients is significantly higher than that of the normal control group,which means the mutation of MTHFR gene may be closely related to the susceptibility of PACG.
