Objective:to screen the differential expression of lincRNA in pancreatic ductal adenocarcinoma and to construct a prognostic model for pancreatic ductal adenocarcinoma,and to provide a reference for the evaluation of the prognosis of patients.Methods:The lincRNA transcriptional data of 146 cases of pancreatic ductal adenocarcinoma and 4 para cancerous tissue lincRNA were downloaded from the American Cancer Genome.First,the differential expression of lincRNA was screened by R software,then the patients with pancreatic ductal adenocarcinoma with incomplete clinical data and less than one month's total survival were removed,and 128 cases of pancreatic ductal adenocarcinoma were finally included.A single factor proportional hazards regression model was used to screen lincRNA which was closely related to the survival time of pancreatic ductal adenocarcinoma.Then 128 patients with pancreatic ductal adenocarcinoma were divided into the training group and the testing group in 64 cases according to the principle of random distribution.The multiple factor Cox regression model was used in the training group to construct a prognostic model with lincRNA related to the survival time of pancreatic ductal adenocarcinoma,and then the pancreatic ducts were selected by using the Akaike information criterion.The best prognosis model of adenocarcinoma was divided into the high risk group and the low risk group with the median value of the prognosis model in the training set.The difference between the high risk group and the low risk group in the training group and the testing group was different when the overall survival rate of the corresponding low risk group was different,which meant the prognosis model of pancreatic duct adenocarcinoma is established.Then using the online web page multi test matrix to predict the target gene of the l:incRNA in the prognosis model,and use the String online web page to draw the protein network interaction map of the target gene,and use the David software to analyze the accumulation of gene ontology and the Encyclopedia of the Kyoto gene and genome.In addition,samples of 20 patients with pancreatic ductal adenocarcinoma were collected,and the expression of lincRNA in the model was verified,and the prognostic value of pancreatic ductal adenocarcinoma prognostic model in the collection of people was also verified.Results:After differential analysis,44 differentially expressed lincRNA were screened out,3 of which were upregulated,and 41 were down regulated.8 of them were associated with the survival of patients with pancreatic ductal adenocarcinoma.The prognosis model of pancreatic ductal adenocarcinoma was =-0.399 ×(RP11-147L13.15 expression)-0.262×(RP11-452H21.4 expression).The prognosis model showed that the overall survival rate of the high risk group in the training group(p=0.009),the testing group(p?0.035),the total population(p=0.001),the male group(p?0.035),the female group(p=0.021),the age older than 65 years(p=0.000),the age less than the 65 year group(p=0.000),was lower than the low risk group.In addition,the prognosis model of pancreatic ductal adenocarcinoma was 0.738,0.754,0.762,0.729,0.753,0.754 and 0.756,respectively,in the training group,the verifying group,the total population,the male group,the female group,the age older than 65 years old and the 5 year survival rate of the 65 years old and equal.The most relevant target gene of RP11-147L13.15 is TRIM52,and the most relevant target gene of RP11-452H21.4 is GAB2.in addition,the top there of GO term that was closely related with the target genes were enriched area of transition metal ion binding(p=0.000),zinc ion binding(p=0.000)and nuclear(p=0.002).In the pathway,only the sphingolipid signaling pathway(p=0.031)was closely related to target genes.The expression of 20 cases of pancreatic ductal adenocarcinoma in RP11-147L13.15 and RP11-452H21.4 were lower than the expression in cancer tissue(P<0.001),high risk group of pancreatic cancer in the general population(p=0.013),male group(p?0.019)and female group(P?0.020),the group more than 65 years old(p=0.046)and less than or equal to the age group of 65 years old(p=0.008)had low overall survival rate than that in the low risk group,and diagnosis of 5 year survival rate of the model for the general population,male group,female group,older than 65 years of age group and age less than or equal to 65 years old group of pancreatic ductal adenocarcinoma patients were respectively 0.681,0.762,0.785,0.642 and 0.966.Conclusion:1.the prognostic model of pancreatic ductal adenocarcinoma lincRNA(RP11-147L13.15 and RP11-452H21.4)is of certain clinical significance for evaluating the prognosis of pancreatic ductal adenocarcinoma patients.2.lincRNA prognostic model for pancreatic ductal adenocarcinoma(RP11-147L13.15 and RP11-452H21.4)is an independent prognostic factor for pancreatic ductal adenocarcinoma...
