Objective:To investigate the effect of iodine on differentiated thyroid cancer and whether iodine and TSH have synergistic effects on differentiated thyroid cancer.Methods:In vitro cell culture differentiated thyroid cancer cells-human thyroid papillary carcinoma cells(TPC-1),according to different concentration of the iodide in the medium,The cells were cultured in different groups:0 mol/l,10-1 mol/l,10-2 mol/l,10-3 mol/l,10-4 mol/l and 10-5 mol/l.And then the proliferation,migration and apoptosis of the cells were detected by the methods like:SRB,Transwell,Scratch assay,and Western blotting.After that,we culture TPC-1 cells by containing different iodine,which is 0 mol/l and 10-3 mol/l.And then the two grous were further divided into four groups according to the differnet TSH concentrations of 0 Um/l and 3 Um/l for culture.Then the cell apoptosis and migration ability were detected by TUNNEL,Transwell,Western blotting and other methods.Results:In the experiment of the effect of iodine on differentiated thyroid cancer.We found that in the concentration gradient of 0~10-1mol/l,the ability of proliferation,migration and resistance of TPC-1 cells increases with the increase of iodine concentration.The apoptotic capacity increases and reaches the highest at 10-3 mol/l.While the iodine concentration further increases,the proliferation,migration and anti-apoptosis ability of TPC-1 cells decrease.Meanwhile,the expression of EMT(epithelial mesenchymal)related protein is also increased.In the experiment of whether iodine and TSH have a synergistic effect.We found that TPC-1 cells in the medium containing both iodine and TSH had the strongest migratory and anti-apoptosis ability compared with the other groups.Without both of the iodine and TSH,TPC-1 cells have the lowest migration and anti-apoptosis ability compared with other groups.The migration and anti-apoptotic capacity of cells containing only iodine or TSH are intervened.Conclusion:To a certain extent,with the increase of iodine concentration,the proliferation and migration of differentiated thyroid carcinoma will increase,reaching a peak at 10-3 mol/l;Iodine may affect differentiated thyroid cancer through EMT pathway;TSH and iodine have the synergistic effect of promoting the growth of differentiated thyroid cancer.
