Objective:To investigate the effect of vitamin D on the expression of CD4+CD25+FoxP3+regulatory T cells in wheezing diseases.Methods:A total of 127 preschool children with wheezing?wheezing group?were selected in our hospital for pediatric pediatrics,and 50 children?healthy group?were collected for physical examination at the child care clinic during the same period.We use enzyme-linked immunosorbent assay?ELISA?to test plasma levels of 25-?OH?D3 and levels of IL-10,IL-4 and TGF-?1,and flow cytometry to test CD4+CD25+FoxP3+regulatory T cells.There were 69 cases of vitamin D insufficient or deficient in children with wheezing.They were the subjects of the study and were divided into two groups according to whether they were supplemented with vitamin D:the control group?37 cases?:conventional therapy,such as anti-infection,anti-tussive,antispasmodic asthma.Vitamin D group?32 cases?:conventional treatment and oral vitamin D treatment for one month.Then we recorded the clinical effects of vitamin D treatment.Results:?1?The levels of serum 25-?OH?D3 in wheezing group were significantly lower than healthy group?P<0.01?;In wheezing group,the proportion of CD4+CD25+FoxP3+regulatory T cells was significantly lower than control group?P<0.01?;Serum concentrations of IL-10 and TGF-?1 in wheezing group were significantly lower than healthy group?P<0.05?;serum level of IL-4 in wheezing group were higher than healthy group?P<0.05?.?2?Before treatment,25-?OH?D3 level,ratio of peripheral blood CD4+CD25+FoxP3+regulatory T cells,IL-10,IL-4,TGF-?1 levels had no significant difference between vitamin D group and the control group,?P>0.05?.After treatment,in the vitamin D group,25-?OH?D3 levels,ratio of CD4+CD25+FoxP3+regulatory T cells,IL-10 and TGF-?1 were significantly higher than before treatment?P<0.05?and IL-4 was significantly lower?P<0.05?.After treatment,in the vitamin D group,25-?OH?D3 levels were significantly higher than control group?P<0.05?,CD4+CD25+FoxP3+regulatory T cells,IL-10,and TGF-?1were increased than control group?P<0.05?,the level of IL-4 cytokine were reduced,however,the result were not statistically significant?P>0.05?.?3?The disappearance time of cough and the length of hospital stay in the vitamin D group were shorter than those in the control group?P<0.05?.Conclusion:?1?Serum levels of 25-?OH?D3 and peripheral blood CD4+CD25+Fox P3+regulatory T cells were decreased in preschool children with wheezing,suggesting a certain degree of vitamin D insufficiency in wheezing diseases.CD4+CD25+Fox P3+regulatory T cells may play a role in the pathogenesis of wheezing diseases;?2?The proportion of CD4+CD25+FoxP3+regulatory T cells in peripheral blood of preschool children with wheezing after vitamin D supplementation is significantly higher,and the clinical effect is obvious.The mechanism may be that vitamin D promotes CD4+CD25+FoxP3+regulatory T cells regulation in peripheral blood.The expression of T cells in turn reduces clinical symptoms. |
