Objective To observe the effect of PRP on photoaging skin of mice by measuring dermis thickness,histological and appearance changes and the expression of type ? and type ? collagen,TGF-?,MMP-1 in the skin of mice after PRP injection.Methods Skin photoaging model was established by irradiating the skin of the back of rats with UV light.Group A was injected with activated PRP(A-PRP);Group B was injected with inactive PRP(N-PRP);Group C was injected with normal saline;Group D was irradiated only with UV;Group E Only shave the hair.After injection for 4 weeks,the appearance and histological characteristics of the rat's skin were evaluated,and the content of collagen type ? and ? in rat skin was semi-quantitatively analyzed by immunohistochemistry.The expression of TGF-? and MMP-1 in rat skin was determined by Western blot.Results Compared with group C and group D,the wrinkles and skin color in group A and group B were significantly improved,and all groups did not return to the level of group E.Histological observation showed that the histological features of group A and group B were significantly better than that of group C and D(P<0.05).The thickness of dermis in group A was higher than that in group B(P<0.05).There was no significant difference between group C and D(P>0.05).The thickness of dermis in group E was significantly higher than that in group C(P<0.01).The expression of type ? collagen in group A and group B was significantly higher than that in group C and D(P<0.01),and the level of collagen ? in group A was higher than that in group B(P<0.05).The expression of type ? collagen in group A and B was significantly higher than that in group C and D(P<0.05),and there was no significant difference between A and B groups(P>0.05).The expression of TGF-? in group A and group B was significantly higher than that in group C and D(P<0.01),the expression of MMP-1 was significantly lower in group A and group B than in group C and D(P<0.01).The expression of TGF-? in group A was higher than that in group B(P<0.05),while the expression of MMP-1 in group A and B had no statistical significance(P>0.05).Conclusion Injection of PRP can improve the appearance and histological features of photo-aged skin in rats.Injection of PRP also can enhance the expression of type ?,type? collagen and TGF-? and decrease the expression of MMP-1 in the photo-aged rat skin.The effect of A-PRP on the expression of type ? collagen and TGF-? is better than that of N-PRP.
