Abstract/Summary:
Objective Colorectal cancer is a colorectal epithelium tumor.It can penetrate the mucosal layer and infiltrate into mucosal substrate.The occurrence of sporadic colorectal cancer is a multi-stage process.Such process contains activation of oncogenes,inactivation of APC and mismatch repair genes,etc.The death rate of patients with sporadic colorectal cancer is obviously higher than that with polyps.Novel candidate gene CAPN1 of s CRC is determined through screening in whole genome.CAPN1 is a member of the calpain.Calpain involves in several basic physiological process,including the remodeling of cytoskeleton,cell signaling transduction,apoptosis and survival.In this study,si RNA is used for the silence of CAPN1 gene,establishing cell model,carrying out cell function experiments and related experiments of RNA and protein level,and determining the correlation between the expression of CAPN1 and the malignant phenotype of colorectal cancer,which proposes new idea for early warning and targeted therapy for s CRC.Methods 1.The expression of CAPN1 in several colorectal cancer cells and normal cells were detected using real-time quantitative PCR and western blotting.2.The knock down efficiency of CAPN1 specificsi RNA,quantitative PCR and western blotting.3.The impact of the loss of CAPN1 on the biological property of CRC cells was investigated in vitro.Results Through quantitative PCR and western blotting,CAPN1 expression is expressed both in colorectal cancer cells and normal cells,but the expression in the colorectal cancer is higher than that in the normal colonal cells.The expression of HT-29 cells is highest,and the expression of Lo Vo is lowest.After inhibition of CAPN1,the declining in expression quantity of CAPN1 in HT-29 can weaken the cell proliferation ability,and affect the migration ability of cells,but the impact on HT-29 cells is not as significant as that on the Lo Vo cells.Conclusion Through in vitro function experiment,it is determined that CAPN1 gene promotes proliferation,colony formation and migration of CRC cells,and CAPN1 probably intervene in the generation and development of sporadic colorectal cancer as an oncogene.
