Background:Gastric cancer?GC?is one of the most common malignant tumors in the digestive tract.Studies have showed that long non-codong RNA?LncRNA?was closely related to the occurrence,development,invasion and metastasis of GC.The present study is to investigate the expression of xloc004787 in GC and its effect on the biological behavior of GC cells.Methods:The expression of xloc004787 was detected by real-time fluorescence quantitative PCR in GC tissues and adjacent tissues?n=52?,and its correlation with clinicopathological features was analyzed.The xloc 004787 overexpression of GC cell lines was studied by using the technique of lentivirus infection,and the changes of biological behavior of GC cells were observed by CCK8,colony formation,scratch test,migration and invasion experiments.Western-blot method was used to observe the expression changes of its related proteins.Results:Real-time fluorescence quantitative PCR results showed that the expression of xloc 004787 in GC was down regulated,and correlated with the depth of tumor invasion?P=0.039?,lymph node metastasis?P=0.002?,TNM stage?P=0.006?and?-catenin protein expression?P=0.048?.However,there was no correlation with age,gender,tumor differentiation,distant metastasis and other clinical pathological parameters?P>0.05?.After over-expression of lentivirus xloc004787 infection,the proliferation,migration and invasion of GC cells were significantly decreased,and the expression of E-cadherin protein increased,and the expression of ?-catenin,N-cadherin,Vimentin,Cyclind1 and cMyc decreased.In vivo,the growth and the volume of tumor xenografts were significantly inhibited and reduced in the lenti-xloc004787 groups compared with the lenti-NC groups.The Ki-67 expression of cell proliferation marker protein also decreased in lenti-xloc004787 groups.Conclusion:In summary,the expression of xloc 004787 is down regulated and involved in the development of GC.It may be expected to become a new biological marker,and provide a new strategy for the diagnosis and treatment of early GC.
